【摘要】:【研究目的】以人骨關(guān)節(jié)炎軟骨標(biāo)本研究對(duì)象,檢測(cè)沉默信息調(diào)節(jié)因子2(SIRT2)在人軟骨組織及細(xì)胞中的表達(dá)分布及表達(dá)情況,探討SIRT2在骨關(guān)節(jié)炎(OA)與正常軟骨組織及細(xì)胞中表達(dá)的差異性!狙芯糠椒ā窟x取臨床中因膝骨關(guān)節(jié)炎而行全膝關(guān)節(jié)置換術(shù)患者的關(guān)節(jié)軟骨標(biāo)本20例,為OA組,按照2007年最新骨關(guān)節(jié)炎診治指南的OA診斷標(biāo)準(zhǔn),所有病人術(shù)前詳細(xì)詢問(wèn)病史,無(wú)外傷、炎癥、關(guān)節(jié)不穩(wěn)定、慢性反復(fù)勞損或先天性疾病等。選取臨床中因股骨頸骨折而行全髖關(guān)節(jié)置換術(shù)患者的關(guān)節(jié)軟骨標(biāo)本10例,為對(duì)照組,患者既往體健,否認(rèn)既往存在關(guān)節(jié)疾病病史,此次股骨頸骨折根據(jù)患者病史及影像學(xué)檢測(cè)排除OA。對(duì)取材關(guān)節(jié)軟骨行免疫熒光染色檢測(cè)SIRT2在兩組軟骨組織的表達(dá)分布及表達(dá)情況。在無(wú)菌條件下,分離軟骨細(xì)胞并進(jìn)行細(xì)胞傳代培養(yǎng),甲苯胺藍(lán)染色進(jìn)行細(xì)胞鑒定。采用Western blot法分別檢測(cè)軟骨組織及細(xì)胞中SIRT2的表達(dá)情況!狙芯拷Y(jié)果】免疫熒光染色結(jié)果顯示SIRT2表達(dá)于軟骨組織中,分布在細(xì)胞核與細(xì)胞質(zhì)中,OA組SIRT2陽(yáng)性細(xì)胞率較對(duì)照組明顯降低(P0.01)。甲苯胺藍(lán)染色后倒置顯微鏡下可見(jiàn):大部分細(xì)胞呈現(xiàn)長(zhǎng)梭形和多角形兩種形態(tài),細(xì)胞長(zhǎng)滿后排列緊密,細(xì)胞核為圓形或者橢圓形,位于胞體中心,細(xì)胞外基質(zhì)均勻,被染成異染的藍(lán)紫色,證實(shí)確為本實(shí)驗(yàn)所需關(guān)節(jié)軟骨細(xì)胞。Western blot結(jié)果顯示OA組軟骨組織中SIRT2表達(dá)水平顯著低于對(duì)照組(P0.01),軟骨細(xì)胞中SIRT2的表達(dá)水平亦顯著低于對(duì)照組(P0.01)。【結(jié)論】SIRT2在人類軟骨組織中有表達(dá),具體表達(dá)在細(xì)胞核與細(xì)胞質(zhì)中,SIRT2表達(dá)降低可能導(dǎo)致骨關(guān)節(jié)炎的發(fā)生及進(jìn)展。
[Abstract]:[objective] to detect the expression distribution and expression of silencing signaling factor 2sirt2 in human cartilage tissue and cells, using human osteoarthritis cartilage specimens. To investigate the difference of expression of SIRT2 in osteoarthritis and normal cartilage tissue and cell. [methods] 20 cases of osteoarthritis of knee joint underwent total knee arthroplasty were selected as OA group. In accordance with the OA diagnostic criteria of the latest guidelines for the diagnosis and treatment of osteoarthritis in 2007, all patients were asked about the history of the disease in detail before operation, without trauma, inflammation, instability of the joint. Chronic recurrent strain or congenital diseases. Ten articular cartilage specimens from patients undergoing total hip arthroplasty due to femoral neck fracture were selected as the control group. The patients were healthy and denied the existence of a history of joint disease. The femoral neck fracture was excluded according to the patient's history and imaging examination. The distribution and expression of SIRT2 in cartilage tissue of the two groups were detected by immunofluorescence staining. The chondrocytes were isolated and cultured. Toluidine blue staining was used to detect the expression of SIRT2 in chondrocytes and chondrocytes by Western blot method. [results] Immunofluorescence staining showed that SIRT2 was expressed in chondrocytes. The percentage of SIRT2 positive cells in OA group was significantly lower than that in control group (P 0.01). After toluidine blue staining, it was found that most of the cells showed long spindle shape and polygonal shape. The nucleus is round or elliptical, located at the center of the cell body, homogenous in the extracellular matrix, and dyed as a blue-purple heterochromatic. The results of Western blot showed that the expression of SIRT2 and SIRT2 in chondrocytes were significantly lower in OA group than in control group. [conclusion] SIRT2 was expressed in human chondrocytes. The decrease of SIRT2 expression in nucleus and cytoplasm may lead to the occurrence and progression of osteoarthritis.
