【摘要】：目的:通過觀測大鼠行結(jié)腸大部切除術(shù)后剩余結(jié)腸中水通道蛋白(Aquaporin AQP)3、8的表達情況及藥物對其的影響,研究術(shù)后剩余結(jié)腸是否有代償性增加水通道蛋白3、8的表達以增強結(jié)腸水分吸收的作用,并探討痛瀉要方、鹽酸洛哌丁胺的止瀉作用是否與促進水通道蛋白3、8的表達有關(guān),從而為臨床上結(jié)腸切除術(shù)后發(fā)生腹瀉提供診療依據(jù)和方法。方法:選取成年雄性SD大鼠36只,按照隨機分組原則分為空白組(A組)、模型組(B組)、痛瀉要方干預(yù)實驗組(C組)、鹽酸洛哌丁胺干預(yù)實驗組(D組),每組9只。其中,A組大鼠單籠常規(guī)飼養(yǎng);B-D組在全麻下行結(jié)腸大部切除端端吻合術(shù),術(shù)后均單籠常規(guī)飼養(yǎng),予抗感染治療3天;C組、D組于術(shù)后第一天起分別予痛瀉要方、鹽酸洛哌丁胺灌胃1周。每組分別于術(shù)后第1、2、4周隨機取3只大鼠結(jié)腸吻合口處腸道組織,A組取臨近手術(shù)部位腸道組織。再按照免疫組化學(xué)和實時熒光定量聚合酶鏈式反應(yīng)(Real-Time RT-PCR)的方法及要求對結(jié)腸組織中AQP3、8的表達進行檢測,最后用統(tǒng)計學(xué)的方法對檢測結(jié)果進行比較。結(jié)果:1.與A組相比,B組、C組、D組大鼠剩余結(jié)腸組織AQP3、8表達均上調(diào),差異有統(tǒng)計學(xué)意義(p0.01)。2.與B組相比,C組、D組大鼠剩余結(jié)腸組織AQP3、8表達含量均上調(diào),差異有統(tǒng)計學(xué)意義(p0.05)。3.與D組相比,C組大鼠剩余結(jié)腸組織AQP3、8表達含量均上調(diào),差異有統(tǒng)計學(xué)意義(p0.05)。結(jié)論:大鼠結(jié)腸大部切除術(shù)后剩余結(jié)腸AQP3、8的表達上調(diào),且其活躍程度與術(shù)后時間有相關(guān)性;痛瀉要方、鹽酸洛哌丁胺對結(jié)腸大部切除術(shù)后剩余結(jié)腸AQP3、8的表達有明顯的促進作用,且痛瀉要方的促進作用較鹽酸洛哌丁胺更明顯。
[Abstract]:Objective: to observe the expression of aquaporin aquaporin aquaporin (Aquaporin) in the residual colon of rats after subtotal colectomy and the effect of drugs on it. To study whether there is a compensatory increase in the expression of aquaporin 3O8 in the residual colon after operation to enhance the water absorption of colon, and to explore whether the antidiarrhea effect of Lupibutylamine Hydrochloride is related to the promotion of the expression of aquaporin 3O8. Methods: 36 adult male Sprague-Dawley rats were selected. According to the principle of random grouping, they were divided into blank group A group, model group B group, Tongxie Yaofang intervention group C group, and loperamine hydrochloride intervention group D group, 9 rats in each group. Descending subtotal resection of colon end-to-end anastomosis, All the patients were fed with single cage after operation and treated with anti-infective therapy for 3 days. Group D was given Tongxie decoction from the first day after operation. Each group was randomly selected from intestinal tissue of colon anastomosis site of 3 rats in 4 weeks after operation. The intestinal tissue of group A was taken from adjacent site of operation. Then according to immunohistochemistry and real-time fluorescence quantitative polymerase, the intestinal tissue of each group was obtained by intragastric administration of lopipramine hydrochloride for 1 week. The method of chain reaction Real-Time RT-PCR was used to detect the expression of AQP3p8 in colonic tissue. Finally, the results were compared with those in group A. results compared with group A, the expression of AQP3p8 in the residual colon of group B and C was up-regulated, and the expression of AQP3p8 in group D was up-regulated. Compared with group B, the expression of AQP3O8 in group D and group C were all up-regulated, and the difference was statistically significant (P < 0.05). Compared with group D, the expression of AQP3O8 in residual colon of group C was up-regulated, and that of group C was significantly higher than that of group C (P < 0.05). Conclusion: after subtotal colectomy, the expression of AQP3O8 in the residual colon of rats is up-regulated, and its activity is correlated with the postoperative time. Lupibutylamine hydrochloride promoted the expression of AQP3O8 in residual colon after subtotal colectomy, and the promoting effect of Tongxieyao recipe was more obvious than that of Lupibutylamine hydrochloride.
