【摘要】：背景:骨橋蛋白是一種細胞外多功能基質糖蛋白,已被證實參與調控骨關節(jié)炎軟骨細胞外基質成分的合成分解代謝,但對于骨關節(jié)炎軟骨細胞本身增殖能力的影響,報道較少。目的:觀察骨橋蛋白對人膝骨關節(jié)炎軟骨細胞增殖能力的影響。方法:所有軟骨標本均取自湘雅醫(yī)院于2012年1至6月因骨關節(jié)炎行膝關節(jié)置換患者,分離培養(yǎng)骨關節(jié)炎軟骨細胞,分別予以骨橋蛋白干預、骨橋蛋白sh RNA慢病毒轉染、空白對照及Con-shR NA轉染等處理,利用MTT及BrdU法檢測細胞的增殖能力變化。結果與結論:(1)骨橋蛋白shR NA慢病毒對骨關節(jié)炎軟骨細胞轉染率為80%;(2)與對照組相比,骨橋蛋白干預可明顯上調骨關節(jié)炎軟骨細胞吸光度值(P0.05),而骨橋蛋白sh RNA慢病毒干擾組吸光度值則顯著下降(P0.05);(3)骨橋蛋白shR NA轉染后,骨橋蛋白的表達水平顯著下降(P0.05);(4)結果表明骨橋蛋白可促進骨關節(jié)炎軟骨細胞增殖,有望成為治療骨關節(jié)炎的新靶點。
[Abstract]:Background: osteopontin is an extracellular multifunctional matrix glycoprotein, which has been proved to be involved in the regulation of the synthesis and catabolism of extracellular matrix components in osteoarthritis chondrocytes, but the effect of osteopontin on the proliferation of osteoarthritis chondrocytes. Objective: to observe the effect of osteopontin on the proliferation of chondrocytes in human knee osteoarthritis. Methods: all cartilage samples were collected from patients undergoing knee arthroplasty in Xiangya Hospital from 2012 to June. Osteoarthritis chondrocytes were isolated and cultured and treated with osteopontin intervention, osteopontin sh RNA lentivirus transfection, blank control and Con-shR na transfection. Results and conclusion the transfection rate of osteopontin shR na lentivirus to osteoarthritis chondrocytes was 802) compared with the control group. Osteopontin intervention significantly upregulated the absorbance of osteoarthritis chondrocytes and decreased the absorbance value of osteopontin sh RNA lentivirus interference group after transfection of osteopontin shR na. The results showed that osteopontin could promote the proliferation of osteoarthritis chondrocytes and might become a new target for the treatment of osteoarthritis.
