雙皮質(zhì)核素鈣調(diào)節(jié)素依賴激酶-1及Ki-67在胰部分切除術(shù)后的表達(dá)及分布

【摘要】:目的:觀察大鼠胰大部分切除術(shù)后雙皮質(zhì)核素鈣調(diào)素依賴性激酶-1(DCAMKL-1)和Ki-67在胰組織中的表達(dá)及分布,探討胰島再生的分子調(diào)控機(jī)制。方法:35只成年SD大鼠隨機(jī)分為正常對照組(5只)和模型組(30只)。模型組再分為6個亞組,每組5只,手術(shù)切除胰約60%~70%并于術(shù)后1 d、2d、3d、4d、5d、6 d分別取殘存胰,免疫組織化學(xué)PV-9001染色法觀察DCAMKL-1和Ki-67在胰組織內(nèi)的表達(dá)及分布。結(jié)果:DCAMKL-1和Ki-67在正常胰組織有少量表達(dá),表達(dá)主要分布在胰腺導(dǎo)管、胰島和腺泡之間的間質(zhì)。模型組術(shù)后1 d,主胰管周圍可見大量細(xì)胞增殖呈蜂巢狀樣排列,DCAMKL-1和Ki-67表達(dá)顯著增高,3 d時主導(dǎo)管周圍增殖的細(xì)胞明顯增多并在其周圍形成較多的小導(dǎo)管,DCAMKL-1和Ki-67表達(dá)于其上皮并達(dá)到頂峰,4 d后開始下降,但至6 d時仍高于正常。與正常對照組相比,DCAMKL-1在胰腺腺泡及胰島內(nèi)的表達(dá)變化不明顯,而Ki-67術(shù)后5 d時達(dá)到高峰,6 d后開始下降。結(jié)論:胰部分切除后胰腺再生中,DCAMKL-1主要表達(dá)在胰主導(dǎo)管上皮,DCAMKL-1+細(xì)胞在主胰管周圍增殖并形成新生導(dǎo)管,DCAMKL-1+細(xì)胞可能為胰干細(xì)胞,DCAMKL-1可能是成體胰腺干細(xì)胞的表面標(biāo)志。
[Abstract]:Objective: to investigate the expression and distribution of Ki-67 and calmodulin-dependent kinase (DCAMKL-1) in rat pancreatic tissue after partial pancreatectomy. Methods 35 adult SD rats were randomly divided into normal control group (n = 5) and model group (n = 30). The model group was subdivided into 6 subgroups with 5 rats in each group. About 60% of the patients underwent surgical resection of the pancreas, and the residual pancreas was removed at 1 day, 2 days, 3 days, 4 days, 5 days and 6 days, respectively. Immunohistochemical PV-9001 staining was used to observe the expression and distribution of DCAMKL-1 and Ki-67 in pancreatic tissues. There was a little expression of DCAMKL-1 and Ki-67 in normal pancreatic tissues. The expression was mainly distributed in the pancreatic duct, the interstitial between the pancreatic islets and the acinus. 1 day after operation, a large number of cells proliferated around the main pancreatic duct in the model group. The expression of DCAMKL-1 and Ki-67 increased significantly at the 3rd day, the number of proliferating cells around the dominant tube increased significantly and more small ducts were formed around them. The expression of DCAMKL-1 and Ki-67 began to decrease 4 days after reaching the peak of the epithelium, but it was still higher than normal at 6 days, compared with the normal control group. The expression of DCAMKL-1 in pancreatic acinar and islets showed no significant change, but reached the peak at 5 d after Ki-67 and began to decrease 6 d after Ki-67. Conclusion: pancreatic regeneration after partial pancreatectomy. DCAMKL-1 was mainly expressed in DCAMKL-1 cells, which proliferated around the main pancreatic duct and formed new ducts. DCAMKL-1 cells may be pancreatic stem cells. DCAMKL-1 may be a surface marker of adult pancreatic stem cells.
