過(guò)氧化物酶體增殖物PGC-1α對(duì)肝缺血再灌注損傷的保護(hù)作用研究

【摘要】:目的探討過(guò)氧化物酶體增殖物受體γ共激活因子-1α(PGC-1α)對(duì)肝缺血再灌注損傷的保護(hù)作用。方法構(gòu)建大鼠肝缺血再灌注損傷模型,恢復(fù)灌注后12h,以蛋白免疫印跡(Western blot)檢測(cè)PGC-1α表達(dá)情況,并檢測(cè)肝臟活性氧、三磷酸腺苷(ATP)水平及血丙氨酸氨基轉(zhuǎn)移酶(ALT)活性變化,評(píng)估肝功能情況。另一方面,構(gòu)建PGC-1α慢病毒過(guò)表達(dá)載體,并在大鼠缺血再灌注前轉(zhuǎn)染,于缺血再灌注后,再以Western blot檢測(cè)PGC-1α表達(dá),肝臟活性氧、ATP水平和血肝酶活性變化,評(píng)估PGC-1α表達(dá)對(duì)肝缺血再灌注損傷的保護(hù)作用。結(jié)果缺血再灌注肝臟PGC-1α表達(dá)較假手術(shù)組及對(duì)照組(未手術(shù))明顯降低(均P0.05),同時(shí)肝臟活性氧族水平及ALT活性顯著升高,而肝ATP產(chǎn)生減少(均P0.05)。PGC-1α慢病毒過(guò)表達(dá)載體轉(zhuǎn)染顯著上調(diào)缺血再灌注肝臟PGC-1α表達(dá),同時(shí)肝臟活性氧族水平及ALT活性較未轉(zhuǎn)染組顯著降低,而肝ATP產(chǎn)生增加(均P0.05)。結(jié)論 PGC-1α表達(dá)有利于保護(hù)肝缺血再灌注損傷。
[Abstract]:Objective to investigate the protective effect of peroxisome proliferator receptor 緯 co-activator 1 偽 (PGC-1 偽) on hepatic ischemia-reperfusion injury in rats. The expression of PGC-1 偽 and reactive oxygen species (Ros) in liver were detected by Western blot 12 hours after reperfusion. Adenosine triphosphate (ATP) level and alanine aminotransferase (alt) activity were measured to evaluate liver function. On the other hand, PGC-1 偽 lentivirus overexpression vector was constructed. The expression of PGC-1 偽, the level of reactive oxygen species (Ros) in liver and the activity of liver enzyme were detected by Western blot after ischemia-reperfusion in rats. To evaluate the protective effect of PGC-1 偽 expression on hepatic ischemia-reperfusion injury. Results the expression of PGC-1 偽 in ischemia-reperfusion liver was significantly lower than that in sham operation group and control group (all P 0.05). At the same time, the level of reactive oxygen species and the activity of ALT in liver were significantly increased. However, liver ATP production was decreased (all P0.05N. PGC-1 偽 lentivirus overexpression vector transfection significantly upregulated liver PGC-1 偽 expression after ischemia reperfusion. At the same time, the level of reactive oxygen species and the activity of ALT in the liver were significantly lower than those in the untransfected group, while the liver ATP production was increased (P 0.05). Conclusion the expression of PGC-1 偽 is beneficial to the protection of liver ischemia-reperfusion injury.
