脈沖射頻大鼠CCI模型背根神經(jīng)節(jié)對脊髓小膠質(zhì)細胞IRF8表達的影響

【摘要】:【目的】本研究選用結(jié)扎大鼠一側(cè)坐骨神經(jīng)主干制備慢性坐骨神經(jīng)縮窄損傷(CCI)模型,通過一般行為學(xué)、50%機械刺激縮爪反應(yīng)閾值(50%MWT)、熱輻射刺激縮爪反應(yīng)潛伏期(TWL)觀察判斷對大鼠背根神經(jīng)節(jié)(DRG)進行脈沖射頻(PRF)治療及鞘內(nèi)注射干擾素調(diào)節(jié)因子8(IRF8)反義寡核苷酸是否具有緩解疼痛的作用,并通過Western-Blot、免疫組化(IHC)法檢測大鼠脊髓組織中IRF8、離子鈣接合蛋白Iba1表達來闡述脈沖射頻背根神經(jīng)節(jié)緩解疼痛的可能機制!痉椒ā康谝徊糠置}沖射頻大鼠CCI模型背根神經(jīng)節(jié)對痛閾和脊髓IRF8表達的影響采用大鼠CCI模型,于造模后第7天行背根神經(jīng)節(jié)脈沖射頻治療。造模前,造模后1、3、5、7天,脈沖射頻后1、7、14天,通過觀察一般行為學(xué),50%MWT、TWL檢測大鼠痛覺改變。造模后7天,脈沖射頻后7、14天,Western-Bolt、IHC法檢測IRF8蛋白水平。第二部分鞘內(nèi)注射IRF8反義ODN對大鼠CCI模型痛閾和脊髓IRF8、Iba1表達的影響建立鞘內(nèi)置管和CCI模型,于造模后12h分別進行鞘內(nèi)注射IRF8反義ODN、IRF8錯義ODN、等體積NS,每天1次,共6天。造模前,造模后1、3、5、7、10、12、14天大鼠實施一般行為學(xué)、50%MWT,TWL測定。造模后第7、14天采用Western-Blot法檢測IRF8、Iba1蛋白表達!窘Y(jié)果】1、與Sham組比,CCI組、CCI+PRF組、CCI+NPRF組大鼠于神經(jīng)損傷后右后爪50%MWT和TWL均顯著降低(P0.01),同時脊髓IRF8蛋白表達上調(diào)(P0.05)。脈沖射頻后1、7、14天CCI+PRF組與治療前比較50%MWT和TWL升高(P0.01),并顯著高于CCI組和CCI+NPRF組(P0.01),但仍低于Sham組(P0.01),脊髓IRF8蛋白的表達則相應(yīng)減少(P0.05)。2、與Sham組比較,NS組、MM組大鼠神經(jīng)損傷后右后爪50%MWT和TWL均顯著降低(P0.01),脊髓IRF8、Iba1蛋白表達則增強(P0.05)。AS組與Sham組相比于CCI后第10天起右后爪50%MWT和TWL下降(P0.01),但仍明顯高于MM組、NS組(P0.01)。CCI后第7、14天IRF8、Iba1蛋白表達,AS組較MM組、NS組顯著減弱(P0.05),但仍強于Sham組(P0.05)。AS組第14天IRF8、Iba1蛋白較第7天增強(P0.05)!窘Y(jié)論】1、PRF可以減輕CCI模型痛敏,其機制可能是PRF抑制大鼠脊髓IRF8的表達,從而緩解疼痛。2、IRF8是誘導(dǎo)外周神經(jīng)損傷后痛覺敏化的關(guān)鍵因子,提前給予阻斷IRF8可以緩解神經(jīng)病理性疼痛。

Western-Blot method was used to detect IRF8 and Iba1 protein expression.
