去細(xì)胞組織工程豬肺動(dòng)脈帶瓣管道的生物相容性研究

【摘要】:目的:探討去細(xì)胞組織工程豬肺動(dòng)脈帶瓣管道的生物相容性。方法:①新鮮豬肺動(dòng)脈帶瓣管道組織3份,A組為對(duì)照組,B、C組采用胰蛋白酶+Triton X-100對(duì)新鮮豬肺動(dòng)脈帶瓣管道進(jìn)行去細(xì)胞處理,C組在去細(xì)胞處理后行明膠嵌合再處理;②對(duì)3組瓣膜及管壁組織行大鼠皮下包埋實(shí)驗(yàn),8周后觀察材料的組織相容性和生長(zhǎng)情況;③通過(guò)動(dòng)態(tài)凝血試驗(yàn)、溶血試驗(yàn)、血小板粘附性能試驗(yàn)分析3種組織材料的血液相容性。結(jié)果:與A組相比,B組及C組的炎性細(xì)胞浸潤(rùn)顯著減少,C組僅有輕微組織反應(yīng);B、C組材料組織相容性好,并于8周后有纖維細(xì)胞長(zhǎng)入。相對(duì)A組及B組,C組的瓣膜及管壁組織具有更低的凝血率,OD-凝血時(shí)間曲線下降更緩慢且吸光度高,同時(shí)血小板粘附更少且沒有明顯變形和偽足。結(jié)論:這種去細(xì)胞組織工程豬肺動(dòng)脈帶瓣管道不僅具有極低的免疫原性,而且具有一定的生長(zhǎng)潛能、良好的組織相容性及血液相容性,有可能成為一種新的、理想的組織工程肺動(dòng)脈帶瓣管道支架材料。
