Lys調(diào)節(jié)PLGA納米纖維的體外降解產(chǎn)物酸度的研究

【摘要】:采用向PLGA納米纖維中添加堿性氨基酸——賴(lài)氨酸(Lys)的方法來(lái)降低降解產(chǎn)物的集酸程度.用電紡絲技術(shù)制備載Lys的PLGA/Lys復(fù)合納米纖維,并用SEM、DSC、及力學(xué)測(cè)試等表征手段對(duì)該復(fù)合納米纖維的形貌、熱學(xué)性質(zhì)以及力學(xué)性質(zhì)等方面進(jìn)行表征;研究PLGA/Lys復(fù)合納米纖維中Lys的釋放特性;對(duì)PLGA與PLGA/Lys納米纖維進(jìn)行體外降解實(shí)驗(yàn),評(píng)價(jià)纖維降解過(guò)程中的形貌和降解液pH值;用已降解8周的納米纖維降解液對(duì)血管平滑肌細(xì)胞進(jìn)行培養(yǎng),MTT檢測(cè)細(xì)胞毒性.結(jié)果表明,在電壓為12 kV,注射速率1.0mL/h,溫度23℃,濕度20%,接收距離14 cm時(shí)可以紡出形貌較好的PLGA/Lys復(fù)合納米纖維,Lys含量越多,復(fù)合納米纖維的直徑越細(xì).納米纖維中Lys的含量影響Lys的釋放特性.Lys促進(jìn)PLGA的降解并對(duì)改善其降解產(chǎn)物的酸度有一定的緩解作用,含2%Lys的納米纖維降解8周后可以使降解液pH維持在7.4~6.7之間.降解的酸性產(chǎn)物不利于平滑肌細(xì)胞生長(zhǎng),但含Lys降解液的培養(yǎng)基中細(xì)胞生長(zhǎng)狀態(tài)良好,這可能是由于Lys能減輕酸性降解產(chǎn)物對(duì)細(xì)胞功能的影響,同時(shí)也能為細(xì)胞增殖提供營(yíng)養(yǎng)物質(zhì).
[Abstract]:The alkaline amino acid-lysine (Lys) was added to the PLGA nanofibers to reduce the acid concentration of the degradation products. PLGA/Lys composite nanofibers loaded with Lys were prepared by electrospinning technique. The morphology, thermal properties and mechanical properties of the composite nanofibers were characterized by SEM,DSC, and mechanical tests. The release characteristics of Lys from PLGA/Lys composite nanofibers were studied, and the degradation experiments of PLGA and PLGA/Lys nanofibers in vitro were carried out to evaluate the morphology and pH value of the degradation solution. Vascular smooth muscle cells (VSMCs) were cultured with biodegradable nanofiber solution for 8 weeks. The cytotoxicity of vascular smooth muscle cells (VSMCs) was detected by MTT.
