魚(yú)皮膠原纖維重組構(gòu)建納吸棉的工藝特征

【摘要】:背景:魚(yú)源膠原基醫(yī)用生物材料在熱穩(wěn)定性、體內(nèi)降解穩(wěn)定性以及體內(nèi)材料形態(tài)學(xué)穩(wěn)定性等方面仍存在明顯不足,利用技術(shù)手段提高淡水魚(yú)類(lèi)膠原海綿材料的密度和膠原分子間結(jié)合的緊密程度進(jìn)而提高材料性能以滿(mǎn)足應(yīng)用需要成為拓展魚(yú)源膠原應(yīng)用領(lǐng)域的當(dāng)務(wù)之急。目的:探究魚(yú)皮膠原纖維重組構(gòu)建納吸棉的工藝條件。方法:以羅非魚(yú)魚(yú)皮為原料,在低于膠原蛋白變性溫度的條件下提取酶促溶性膠原蛋白,以膠原蛋白纖維重組率為考察指標(biāo),研究膠原蛋白纖維重組工藝并對(duì)其進(jìn)行參數(shù)優(yōu)化,確定了魚(yú)皮膠原纖維重組構(gòu)建納吸棉的最佳工藝。以此為基礎(chǔ)進(jìn)行重組納吸棉的紅外光譜和微觀結(jié)構(gòu)分析。結(jié)果與結(jié)論:通過(guò)單因素試驗(yàn)和正交試驗(yàn)得出最佳魚(yú)皮膠原纖維重組反應(yīng)條件為:膠原質(zhì)量濃度為1 g/L,離子強(qiáng)度(Na Cl濃度)為65 mmol/L,p H值7.4,體系溫度為20℃,重組時(shí)間為10 h,在該條件下得到的纖維重組率高達(dá)68.6%。在此基礎(chǔ)上制備的納吸棉具有細(xì)線(xiàn)狀纖維構(gòu)成的多孔網(wǎng)狀結(jié)構(gòu),且其三螺旋結(jié)構(gòu)完整,是一種性能優(yōu)良的腔內(nèi)止血材料。
[Abstract]:Background: there are still obvious deficiencies in thermal stability, degradation stability and morphological stability of fish-derived collagen-based biomaterials. It is an urgent task to improve the density of freshwater fish collagen sponge materials and the density of collagen molecules and to improve the properties of the materials to meet the needs of the application of fish collagen. Objective: to explore the technological conditions of constructing nano-absorbent cotton by recombination of collagen fiber from fish skin. Methods: using tilapia skin as raw material, the enzyme-enhanced collagen was extracted under the condition of lower than the denaturation temperature of collagen. The recombination rate of collagen fiber was used as the index to study the technology of collagen fiber recombination and its parameters were optimized. The optimum technology of recombining fish skin collagen fiber to construct nano-absorbent cotton was determined. On the basis of this, the infrared spectrum and microstructure analysis of the reconstituted nano-absorbent cotton were carried out. Results and conclusion: the optimum reaction conditions of fish skin collagen fiber recombination were as follows: collagen concentration was 1 g / L and ionic strength (Na Cl concentration was 65 mmol/L,p H = 7.4, and the optimal reaction conditions were as follows: collagen concentration was 1 g / L and ionic strength concentration was 7.4mol 路L ~ (- 1).
