不同復(fù)溫速率對(duì)含硫酸軟骨素的液氮保存心臟瓣膜的影響

【摘要】:目的比較不同復(fù)溫速率對(duì)液氮保存的同種異體心臟瓣膜的細(xì)胞活性及瓣膜結(jié)構(gòu)的影響。方法將48只兔主動(dòng)脈瓣隨機(jī)分成4組,每組12只,均經(jīng)滅菌處理。對(duì)照組于滅菌后立即行瓣膜細(xì)胞活力測(cè)定及組織學(xué)檢查;實(shí)驗(yàn)Ⅰ~Ⅲ組采用程序控溫降溫法將瓣膜保存在含硫酸軟骨素的液氮深低溫中1個(gè)月后,分別采用100、30、15℃/min的平均復(fù)溫速率進(jìn)行復(fù)溫,復(fù)溫后行瓣膜細(xì)胞活力測(cè)定、光鏡及電鏡檢查。結(jié)果 3種復(fù)溫方法對(duì)瓣膜均有損害,與對(duì)照組比較,實(shí)驗(yàn)Ⅰ組瓣膜組織學(xué)結(jié)構(gòu)損傷較重,細(xì)胞活力下降顯著(P0.05);實(shí)驗(yàn)Ⅱ組瓣膜組織學(xué)結(jié)構(gòu)損傷較輕,細(xì)胞活力下降顯著(P0.05);實(shí)驗(yàn)Ⅲ組瓣膜組織學(xué)結(jié)構(gòu)改變輕微,瓣膜細(xì)胞活力下降不明顯(P0.05)。結(jié)論冷凍保存后將復(fù)溫速率控制在15℃/min能更好地保護(hù)心臟瓣膜,經(jīng)此法復(fù)溫的瓣膜組織學(xué)結(jié)構(gòu)損傷輕微,細(xì)胞活力佳,瓣膜可用性大。
