接枝不同官能團的聚酯材料對血小板黏附及其功能的影響研究

【摘要】:目的考察接枝不同官能團(-COOH、-OH、兩性基團)的聚對苯二甲酸丁二醇酯(PBT)膜對血小板黏附及血小板功能的影響,選擇適用于過濾去除濃縮血小板中白細胞的材料。方法利用紫外改性的方法,將不同單體包括丙烯酸(AA)、丙烯酸羥乙酯(HEA)和N-(3-磺丙基)-N-甲基丙烯酸基氧乙基-N,N-二甲基銨內(nèi)銨鹽(SMDB)接枝到PBT表面,得到接枝不同官能團的PBT膜,包括PBT-AA(-COOH)、PBT-HEA(-OH)、PBT-SMDB(兩性基團);通過傅里葉紅外光譜、水接觸角及表面粗糙度等指標對改性PBT的理化性能做表征,采用掃描電鏡觀察改性PBT對血小板的黏附,并通過檢測PBT改性前后的血小板聚集率、低滲休克相對變化、CD62p表達率等指標,考察改性PBT對血小板功能的影響。結(jié)果在紅外光譜的1 047和1 043 cm-1處出現(xiàn)了C-O-C的彎曲振動峰和O=S=O的對稱伸縮振動峰,提示HEA和SMDB接枝到了PBT膜表面。掃描電鏡顯示:未改性的PBT膜表面黏附的血小板數(shù)明顯多于改性PBT膜表面,且可以觀察到明顯的偽足。血小板黏附數(shù)在接枝不同官能團的PBT膜表面順序為:-COOH兩性基團-OH。未改性PBT組(n=6)與對照組(未接觸材料的血小板)(n=6)比較:血小板最大聚集率(%)為87.22±2.10 vs 82.36±2.11,低滲休克相對變化率(%)為79.33±1.27 vs 77.12±1.09,CD62p表達率增加(%)為9.78±0.58 vs 17.45±1.25(P0.05)。在接枝不同官能團的改性PBT膜中,PBT-HEA(n=6)對血小板聚集和血小板活化等功能影響最小[血小板最大聚集率(93.34±1.45)%、血小板低滲休克相對變化率(81.26±1.48)%、CD62p表達率(12.51±0.85)%]。結(jié)論表面接枝HEA的聚酯對血小板的黏附及功能影響明顯小于接枝其他官能團的材料,或可作為用于濃縮血小板過濾去除白細胞的新材料。
[Abstract]:Objective to investigate the effects of poly (butylene terephthalate) (PBT) membranes grafted with different functional groups (- COOH,-OH,) on platelet adhesion and platelet function. Methods different monomers, including (AA), hydroxyethyl acrylate (HEA) and N- (3-sulfopropyl) -N-methacrylate ethoxyethyl N, were modified by UV. (SMDB) was grafted onto the surface of PBT to obtain PBT membrane grafted with different functional groups, including PBT-AA (- COOH), PBT-HEA (- OH), PBT-SMDB). The physicochemical properties of modified PBT were characterized by FTIR, water contact angle and surface roughness. The adhesion of modified PBT to platelet was observed by scanning electron microscope, and the platelet aggregation rate before and after PBT modification was measured. The effects of modified PBT on platelet function were investigated by relative changes of hypotonic shock and CD62p expression. Results the bending vibration peak of C-O-C and the symmetrical stretching vibration peak of O=S=O appeared at 1 047 and 1 043 cm-1 of IR spectrum, suggesting that HEA and SMDB grafted onto the surface of PBT film. Scanning electron microscopy showed that the number of platelets adhered to the surface of the unmodified PBT membrane was significantly higher than that of the modified PBT membrane, and obvious pseudopodia could be observed. The order of platelet adhesion number on the surface of PBT membrane grafted with different functional groups is-COOH amphoteric group-OH. The maximum platelet aggregation rate (%) was 87.22 鹵2.10 vs (82.36 鹵2.11) in the unmodified PBT group and the control group (non-contact material platelets). The relative change rate of hypotonic shock (%) was 79.33 鹵1.27 vs, 77.12 鹵1.09 vs, the expression rate of CD62p increased (%) was 9.78 鹵0.58 vs 17.45 鹵1.25 (P0.05). Among the modified PBT membranes grafted with different functional groups, PBT-HEA (NN6) had the least effect on platelet aggregation and platelet activation [the maximum platelet aggregation rate was (93.34 鹵1.45)%]. The relative change rate of platelet hypotonic shock was (81.26 鹵1.48)% and the expression rate of CD62p was (12.51 鹵0.85)%. Conclusion the effect of polyester grafted with HEA on platelet adhesion and function is obviously smaller than that on other functional groups, and it may be used as a new material for platelet filtration and leukocyte removal.
