類人膠原蛋白-微孔淀粉的制備及生物學評價

【摘要】:文中以微孔淀粉為主體,研究一種新型復(fù)合止血材料的制備及生物學評價。此復(fù)合材料由一定比例的α-淀粉酶與糖化酶復(fù)合后處理淀粉以形成微孔結(jié)構(gòu),然后復(fù)合類人膠原蛋白,再以戊二醛為交聯(lián)劑進行交聯(lián);用SD大鼠作肝臟出血模型進行止血效果對比試驗。通過正交試驗確定微孔淀粉最優(yōu)制備工藝條件如下:α-淀粉酶:糖化酶=1∶3,T/℃=50,t/h=20,pH=4.8;用0.2%(V/V)戊二醛乙醇溶液交聯(lián)的類人膠原蛋白-微孔淀粉止血材料,在相同的止血時間下,該復(fù)合材料平均吸水率為3 580%,對照組(分別是微孔淀粉和類人膠原蛋白海綿)的平均吸水率為3 160%和3 280%。而在SD大鼠肝臟實驗中,復(fù)合材料t/s=94±13,而外購的止血劑t/s=121±15。類人膠原蛋白-微孔淀粉復(fù)合材料止血的止血原理從3個方面展開:①微孔淀粉吸收水分并將血液中的有形物質(zhì)聚集到表面;②加速內(nèi)源性凝血因子的激活使其形成血栓;③在類人膠原蛋白的牽引下,淀粉顆粒聚成團大幅度增加了比表面積,使止血效果更優(yōu)秀;對照組止血機理只包含了該復(fù)合材料的前兩個方面。結(jié)果顯示,復(fù)合材料的止血效果要優(yōu)于普通微孔淀粉制成的止血劑,證明該復(fù)合止血材料具有良好的止血性能。
