模擬體內(nèi)應(yīng)力作用的骨支架降解和骨重建仿真研究

【摘要】:目前對骨組織工程學(xué)的研究主要通過實(shí)驗(yàn)研究和數(shù)值模擬的方法進(jìn)行。由于骨再生過程持續(xù)時(shí)間周期很長,基于體內(nèi)試驗(yàn)的方法開展相關(guān)研究難于進(jìn)行,且花費(fèi)巨大。隨著計(jì)算機(jī)計(jì)算能力的提高和新的數(shù)學(xué)理論的不斷出現(xiàn),利用數(shù)值模擬技術(shù)對骨組織工程進(jìn)行相關(guān)研究的工作逐步深入。 本論文主要通過計(jì)算機(jī)仿真的手段對體內(nèi)應(yīng)力作用下的骨支架降解和骨重建的過程進(jìn)行模擬研究。主要工作和成果如下: (1)構(gòu)建了考慮應(yīng)力影響因素的骨支架降解公式,利用多孔固體結(jié)構(gòu)與性能理論建立起骨支架的彈性模量和支架壁厚之間的關(guān)系式,描述骨支架胞元的降解,得出每次降解骨支架的壁厚的變化量。 (2)利用應(yīng)變能密度理論模擬伴隨骨支架降解過程的骨再生過程。以參考應(yīng)變能密度理論為基礎(chǔ),在計(jì)算應(yīng)變能時(shí)綜合考慮降解過程中新骨的生成對骨-支架系統(tǒng)的力學(xué)性能的影響,骨支架降解和新骨重建是兩個(gè)獨(dú)立,又相互影響的過程。 (3)應(yīng)用有限元軟件和子程序來模擬骨支架降解和骨再生的過程。利用骨支架胞元的變化刻畫整體支架的變化,把有限元軟件中的六面體單元和骨支架胞元相結(jié)合,以單元的力學(xué)性能的變化過程解釋整個(gè)降解過程的發(fā)生。選取不同骨支架的初始孔徑分別對上述過程進(jìn)行計(jì)算,結(jié)果表明,隨著孔徑在一定范圍內(nèi)的增大,骨重建的情況越理想。 本論文建立的骨支架降解模型考慮了應(yīng)力的影響,能夠?yàn)槭茌^大應(yīng)力作用下的支架降解的情況做更清晰的描述。此外,論文中提出的以力學(xué)性能變化來刻畫幾何變化的仿真方法,在模擬過程中簡化了大量的計(jì)算工作,可為以后相關(guān)的研究提供參考。
[Abstract]:At present, the research of bone tissue engineering is mainly carried out by experimental research and numerical simulation. Due to the long duration of bone regeneration, it is difficult and costly to carry out the research based on in vivo test. With the improvement of computer computing ability and the emergence of new mathematical theory, the research on bone tissue engineering by numerical simulation technology has been deepened step by step. In this paper, the process of bone scaffold degradation and bone reconstruction under stress in vivo was simulated by computer simulation. The main work and achievements are as follows: (1) the degradation formula of bone scaffold considering the factors of stress is constructed. The relationship between elastic modulus and wall thickness of bone scaffold is established by using the theory of porous solid structure and properties. The cell degradation of bone scaffold was described, and the change of wall thickness of bone scaffold was obtained. (2) the strain energy density theory was used to simulate the process of bone regeneration with the degradation of bone scaffolds. Based on the theory of reference strain energy density, the influence of the formation of new bone on the mechanical properties of bone scaffold system is considered synthetically in the calculation of strain energy. The degradation of bone scaffold and the reconstruction of new bone are two independent and interactive processes. (3) finite element software and subroutine were used to simulate the degradation and bone regeneration of bone scaffold. The change of bone scaffold cell is used to describe the change of the whole scaffold. The hexahedron element and bone scaffold cell in finite element software are combined to explain the whole degradation process with the change process of the mechanical properties of the unit. The process was calculated with the initial aperture of different bone scaffolds. The results show that with the increase of the pore diameter in a certain range, the better the situation of bone reconstruction. In this paper, the degradation model of bone scaffold takes into account the effect of stress, which can clearly describe the degradation of scaffold under high stress. In addition, the simulation method proposed in this paper to describe the geometric change with the change of mechanical properties simplifies a lot of calculation work, which can be used as a reference for the related research in the future.
