脫鈣松質(zhì)骨體外構(gòu)建組織工程軟骨的實(shí)驗(yàn)研究

【摘要】:目的軟骨細(xì)胞復(fù)合同種異體脫鈣松質(zhì)骨(demineralized cancellous bone,DCB)體外構(gòu)建組織工程軟骨,評價DCB作為組織工程軟骨支架的可行性。方法取新西蘭兔長骨干骺端松質(zhì)骨制備DCB,掃描電鏡觀察;分離幼兔關(guān)節(jié)軟骨細(xì)胞,原代培養(yǎng)后種植在同種異體DCB體外培養(yǎng),分別于7、14、21、28、35、42d取材進(jìn)行蘇木素-伊紅(HE)、甲苯胺藍(lán)(TB)、Ⅰ型和Ⅱ型膠原染色。結(jié)果制備的DCB成三維立體多孔結(jié)構(gòu),孔隙大小100~500μm,有良好的可塑性和一定的力學(xué)強(qiáng)度;軟骨細(xì)胞在DCB表面和空隙內(nèi)生長、分化良好,形成軟骨陷窩,分泌基質(zhì)蛋白聚糖和Ⅱ型膠原。培養(yǎng)42d軟骨細(xì)胞在DCB表面及孔隙內(nèi)形成軟骨樣組織。結(jié)論 DCB復(fù)合同種異體軟骨細(xì)胞在體外成功構(gòu)建了組織工程軟骨,是一種較理想的軟骨組織工程支架材料。
