【摘要】：目的以巴沙魚皮為原料制備新型膠原支架材料,并對該材料的結(jié)構(gòu)特性、物理性質(zhì)以及體外降解性進(jìn)行檢測和分析,以探索巴沙魚是否可以取代陸生哺乳動物作為膠原支架材料的合適來源。方法巴沙魚皮經(jīng)過反復(fù)脫脂、脫色、脫雜處理后,冷凍干燥成膜狀。采用凱氏定氮法測定粗蛋白含量,掃描電鏡觀察和分析材料的結(jié)構(gòu)以及孔隙的大小和分布,液體置換法測孔隙率,萬能試驗(yàn)機(jī)檢測抗拉強(qiáng)度。觀察材料的黏度隨溫度的變化情況,以檢測材料的變性溫度。將材料浸泡在置于37℃恒溫震蕩箱的磷酸鹽緩沖液(0.1mol/L,pH 7.4)中,檢測材料的吸水率和失重率。結(jié)果材料的粗蛋白含量為95.2%。肉眼觀察厚薄均勻。掃描電鏡顯示材料的一面較為粗糙,可見多孔結(jié)構(gòu),孔徑大小不一,分布均勻;另一面光滑,孔隙致密�？紫堵�(55.50±1.94)%,厚度(0.66±0.10)mm,抗拉強(qiáng)度(18.82±0.94)MPa。未經(jīng)交聯(lián)的材料變性溫度為34℃,經(jīng)熱交聯(lián)后為36℃。48h吸水率(379.77±77.81)%。經(jīng)熱交聯(lián)后的材料28d的失重率為(80.22±2.49)%,緩沖液pH為6.67±0.05。結(jié)論巴沙魚皮膠原支架材料可制成厚薄均勻、具有致密層和疏松層雙層不同結(jié)構(gòu)的生物膜。材料具有良好的機(jī)械強(qiáng)度、較為適宜的變性溫度,但降解速率較快,有待改進(jìn)。
