離子濃度和滲透壓在生物活性玻璃抗菌中的作用

【摘要】:生物材料是一類高新技術(shù)材料,在人體組織、器官的診斷和修復(fù)過程中與天然材料和其它人造材料相比,功能優(yōu)異。生物材料已被廣泛用于治療臨床的各種運動損傷,其開發(fā)經(jīng)歷了三個標(biāo)志性的階段。硅酸鹽生物活性玻璃是第二代生物材料,根據(jù)含硅量不同,分為數(shù)種,其中45S5最具有代表性。 細(xì)菌感染是臨床各種常見手術(shù)中難題之一。在不增加副作用的前提下,如果生物活性玻璃本身具有抗菌性,生物材料在臨床應(yīng)用中將會有較大的進(jìn)展。專家致力于硅酸鹽生物活性玻璃的抗菌研究和應(yīng)用,取得了有意義的突破,證實了其抗菌特性。 文獻(xiàn)報導(dǎo)影響生物活性玻璃抗菌的可能因素有液體環(huán)境的pH值、粉體顆粒粒徑、粉體對細(xì)菌的粘附、液體環(huán)境的離子濃度和滲透壓等,但離子濃度和滲透壓的因素尚未得到進(jìn)一步證實。本文將液體環(huán)境中離子濃度和滲透壓作為研究指標(biāo),探討離子濃度和滲透壓在硅酸鹽生物活性玻璃抗菌中的作用。 方法:測定45S5粉體不同濃度和不同浸提時間的浸提液滲透壓,根據(jù)45S5滲透壓范圍配制更大范圍滲透壓的中性溶液,細(xì)菌培養(yǎng)后分析數(shù)據(jù)得出結(jié)論。測定粉體浸提液的鈉鈣濃度,配制更大范圍離子濃度的中性溶液,細(xì)菌培養(yǎng)后分析數(shù)據(jù)得出結(jié)論。 結(jié)果:1、45S5粉體浸提液的滲透壓值為420-476mosm/kg.不同浸提時間對于滲透壓的影響沒有顯著性影響,不同粉體濃度對滲透壓有顯著性影響;2、滲透壓在200-600mosm/kg范圍的中性溶液內(nèi),細(xì)菌生長無明顯變化;3、Na+濃度為4.41-12.34mg/ml,Ca2+濃度為0.009-0.281mg/ml范圍的中性溶液內(nèi),細(xì)菌生長無明顯變化。 結(jié)論:1、生物活性玻璃粉體浸提液的滲透壓和粉體濃度成正相關(guān),與浸提時間無明顯相關(guān)性。2、細(xì)菌在200-600mosm/kg的液體環(huán)境中生長無明顯變化,即此范圍內(nèi)滲透壓在生物活性玻璃抗菌中無明顯作用3、細(xì)菌在Na+濃度為4.41-12.34mg/ml,Ca2+濃度為0.009-0.281mg/ml的液體環(huán)境中生長無明顯變化仍能存活,即此范圍內(nèi)Na+和Ca2+濃度在生物活性玻璃抗菌中無明顯作用。
[Abstract]:Biomaterials are a kind of high-tech materials, which have excellent function compared with natural materials and other artificial materials in the diagnosis and repair of human tissues and organs. Biomaterials have been widely used in the treatment of various clinical sports injuries, its development has undergone three iconic stages. Silicate biologically active glass is the second generation biomaterial. It can be divided into several kinds according to the content of silicon, among which 45S5 is the most representative. Bacterial infection is one of the common surgical problems in clinic. On the premise of no side effects, if the bioactive glass itself has antibacterial properties, biomaterials will make great progress in clinical application. Experts devoted themselves to the antibacterial research and application of silicate biologically active glass. The possible factors affecting the antibacterial activity of bioactive glass are pH value of liquid environment, particle size, adhesion of powder to bacteria, ionic concentration and osmotic pressure of liquid environment, etc. However, the factors of ion concentration and osmotic pressure have not been further confirmed. In this paper, ionic concentration and osmotic pressure in liquid environment are taken as the research index, and the effect of ion concentration and osmotic pressure on antibacterial activity of silicate bioactive glass is discussed. Methods: the osmotic pressure of 45S5 powder was determined with different concentration and time. The neutral solution with larger osmotic pressure was prepared according to the range of 45S5 osmotic pressure. The concentration of sodium and calcium in the extract of powder was determined, and the neutral solution with larger concentration of ions was prepared. The data of bacteria culture were analyzed and the conclusion was drawn. Results: 1the osmotic pressure of the extract was 420-476mosm / kg. Different extraction time had no significant effect on osmotic pressure, but different concentration of powder had significant effect on osmotic pressure, 2, osmotic pressure in neutral solution of 200-600mosm/kg had no obvious change. In neutral solution with the concentration of 4.41 ~ 12.34 mg / ml Ca ~ (2 +) in the range of 0.009-0.281mg/ml, there was no significant change in bacterial growth. Conclusion: 1. The osmotic pressure of the bioactive glass powder extract was positively correlated with the concentration of the powder, but had no significant correlation with the extraction time. 2. There was no obvious change in the growth of bacteria in the liquid environment of 200-600mosm/kg. In this range, the osmotic pressure had no obvious effect on the antibacterial activity of bioactive glass. The bacteria could survive in the liquid environment where the concentration of Na was 4.41 ~ 12.34 mg / ml / ml Ca ~ (2 +) and the concentration of Ca ~ (2 +) was 0.009-0.281mg/ml. In this range, the concentration of Na and Ca2 had no obvious effect on the antibacterial activity of bioactive glass.
