【摘要】：背景:包裹幽門螺桿菌全菌蛋白抗原的研究仍處于探索階段,有關(guān)殼聚糖微球包裹幽門螺桿菌全菌蛋白抗原的制備工藝及體外釋放性能的文獻(xiàn)甚少。目的:探討幽門螺桿菌全菌蛋白抗原殼聚糖微球的制備工藝及體外釋放特性。方法:采用沉淀法制備殼聚糖微球,篩選最佳制備工藝及配比、包裹時(shí)間,并在電鏡下觀察微球的形態(tài)和粒徑。采用殼聚糖微球包裹幽門螺桿菌全菌蛋白抗原,BCA法測(cè)定幽門螺桿菌全菌蛋白抗原微球的包裹率、包裹量及體外釋放率。結(jié)果與結(jié)論:終體積分?jǐn)?shù)為1%的冰醋酸、硫酸鈉為交聯(lián)劑、pH 5.0、滴加交聯(lián)劑時(shí)不粉碎處理為殼聚糖微球最佳制備工藝,電鏡觀察顯示微球表面光滑、形態(tài)圓整,具有良好的分散性,多數(shù)微球粒徑為1.0-5.0μm。幽門螺桿菌全菌蛋白抗原微球的包裹率為80.4%,包裹量為16.4%,48 h總釋放率為19.4%,幽門螺桿菌全菌蛋白抗原微球整體呈緩慢釋放狀態(tài)。結(jié)果證實(shí),實(shí)驗(yàn)制備的殼聚糖微球?qū)τ拈T螺樣菌全菌蛋白抗原具有良好的包裹率和包裹量,幽門螺桿菌全菌蛋白抗原微球整體呈緩慢釋放狀態(tài)。
[Abstract]:Background: the study of the whole protein antigen of Helicobacter pylori is still in the exploratory stage. There are few literatures about the preparation of the protein antigen of Helicobacter pylori wrapped by chitosan microspheres and its release in vitro. Aim: to study the preparation and release characteristics of Helicobacter pylori protein antigen chitosan microspheres. Methods: chitosan microspheres were prepared by precipitation method. The optimum preparation technology and ratio, the encapsulation time and the morphology and particle size of chitosan microspheres were observed under electron microscope. The encapsulation rate, encapsulation quantity and in vitro release rate of Helicobacter pylori whole protein antigen microspheres were determined by chitosan microspheres. Results and conclusion: 1% final volume fraction of glacial acetic acid, sodium sulfate as crosslinking agent (pH 5.0), no comminution when adding crosslinking agent, was the best preparation technology of chitosan microspheres. Electron microscope observation showed that the surface of microspheres was smooth and the shape was round. The particle size of most microspheres was 1.0-5.0 渭 m. The encapsulation rate of the whole protein antigen microspheres of Helicobacter pylori was 80.4 and the total release rate of the whole protein antigen microspheres of Helicobacter pylori in 48 hours was 19.4.The whole microsphere of Helicobacter pylori protein antigen was slowly released. The results showed that the prepared chitosan microspheres had a good encapsulation rate and encapsulation capacity to the whole protein antigen of Helicobacter pylori, and the whole protein microspheres of Helicobacter pylori showed a slow release state.
