體外評價新型根尖倒充填材料iRoot的生物學(xué)性能

【摘要】:目的:體外評價新型根尖倒充填材料iRoot BP plus和iRoot FS的生物學(xué)性能。方法:(1)將牙根預(yù)備成長3 mm、根管直徑1 mm的試樣,分別于根管內(nèi)填充iRoot BP plus、iRoot FS和三氧化礦物凝聚體(mineral trioxide aggregate,MTA)。將制備好的試樣置于模擬體液(simulated body fluid,SBF)中,用掃描電子顯微鏡觀察暴露的材料表面礦物沉積情況,并通過X射線能譜對其表面形成的晶體進(jìn)行元素分析,測定SBF的p H值隨時間的變化。(2)將iRoot BP plus、iRoot FS和MTA制備成直徑8 mm、高度2 mm的圓柱,制備DMEM浸提液,通過MTT實驗觀察材料對MG63細(xì)胞增殖活性的影響,并通過實時熒光定量PCR和堿性磷酸酶(alkaline phosphatase,ALP)染色分析材料對ALP基因及蛋白表達(dá)的影響。結(jié)果:(1)在SBF中浸泡24 h時,iRoot BP plus、iRoot FS和MTA表面均已有礦物沉積,14 d后有大量礦物沉積,鈣∶磷比值分別為1.43、1.39和1.51;(2)iRoot BP plus、iRoot FS和MTA均可使SBF的p H升高,3周時分別為8.09±0.07、7.91±0.06和8.11±0.06;(3)以體積比為1∶5和1∶10稀釋的iRoot BP plus、iRoot FS和MTA浸提液對MG63細(xì)胞的增殖無明顯促進(jìn)或抑制作用;(4)iRoot BP plus、iRoot FS和MTA組對MG63細(xì)胞ALP基因的表達(dá)均具有促進(jìn)作用,ALP染色各組未見明顯差異。結(jié)論:iRoot在模擬體液中可誘導(dǎo)礦物質(zhì)沉積,促進(jìn)MG63細(xì)胞的分化和礦化,具有良好的生物活性和促成骨作用。
[Abstract]:Objective: to evaluate the biological properties of iRoot BP plus and iRoot FS in vitro. Methods: (1) the specimens with 3 mm root preparation and 1 mm diameter root canal were filled with iRoot BP plus iRoot FS and (mineral trioxide aggregate MTA, respectively. The prepared samples were placed in simulated body fluid (simulated body fluidge). The surface mineral deposits of exposed materials were observed by scanning electron microscope (SEM), and the elements of crystals formed on the surface were analyzed by X-ray spectroscopy. The pH value of SBF was measured with time. (2) the cells were prepared into 8mm diameter and 2mm high cylinder by iRoot BP plusiRoot FS and MTA. The DMEM extract was prepared. The effect of the material on the proliferation of MG63 cells was observed by MTT assay. The effect of alkaline phosphatase on the expression of ALP gene and protein was analyzed by real-time fluorescence quantitative PCR and alkaline phosphatase (alkaline) staining. Results: (1) after immersion in SBF for 24 h, a large number of minerals were deposited on the surfaces of iRoot BP plusiRoot FS and MTA after 14 days. The ratio of calcium to phosphorus was 1.43 鹵1.39 and 1.51 respectively; (2) the pH of SBF was 8.09 鹵0.07 鹵7.91 鹵0.06 and 8.11 鹵0.06 at 3 weeks, respectively; (2) the ratio of iRoot BP plusiRoot FS and MTA diluted at 1:5 and 1:10 did not significantly promote the proliferation of MG63 cells. (4) both iRoot BP plusiRoot FS and MTA groups could promote the expression of ALP gene in MG63 cells. ConclusionThe specific root can induce mineral deposition and promote differentiation and mineralization of MG63 cells in simulated body fluid. It has good biological activity and bone promoting effect.
