同種異體和異種來源骨髓間充質(zhì)干細(xì)胞誘導(dǎo)成軟骨修復(fù)喉軟骨缺損

【摘要】:背景:由于軟骨細(xì)胞缺乏再生能力,選擇合適的種子細(xì)胞是修復(fù)軟骨缺損需首要解決的問題。目的:探討同種異體和異種來源骨髓間充質(zhì)干細(xì)胞成軟骨誘導(dǎo)后修復(fù)喉軟骨缺損的效果。方法:取第3代人骨髓間充質(zhì)干細(xì)胞和兔骨髓間充質(zhì)干細(xì)胞加入軟骨定向誘導(dǎo)液(含轉(zhuǎn)化生長因子β1和骨形態(tài)發(fā)生蛋白)進(jìn)行成軟骨誘導(dǎo),并滴加于聚乳酸-羥基乙酸共聚物支架上。取30只新西蘭大白兔隨機(jī)分為3組:空白對照組、人骨髓間充質(zhì)干細(xì)胞修復(fù)組和兔骨髓間充質(zhì)干細(xì)胞修復(fù)組,制備喉軟骨缺損動物模型,分別植入生理鹽水浸濕的聚乳酸-羥基乙酸共聚物支架,人骨髓間充質(zhì)干細(xì)胞復(fù)合聚乳酸-羥基乙酸共聚物支架,兔骨髓間充質(zhì)干細(xì)胞復(fù)合聚乳酸-羥基乙酸共聚物支架。術(shù)后4周和8周,免疫組化法檢測喉部組織Ⅱ型膠原的表達(dá)。結(jié)果與結(jié)論:各組動物均呼吸通暢、正常,未出現(xiàn)喘鳴等,進(jìn)食和活動情況良好,未出現(xiàn)化膿或者感染現(xiàn)象。術(shù)后4周和8周,人骨髓間充質(zhì)干細(xì)胞修復(fù)組和兔骨髓間充質(zhì)干細(xì)胞修復(fù)組的Ⅱ型膠原陽性率均顯著高于空白對照組(P0.05)。人骨髓間充質(zhì)干細(xì)胞修復(fù)組和兔骨髓間充質(zhì)干細(xì)胞修復(fù)組間比較,差異無顯著性意義(P0.05)。結(jié)果表明同種異體和異種來源的骨髓間充質(zhì)干細(xì)胞成軟骨誘導(dǎo)后進(jìn)行兔喉軟骨缺損修復(fù)均可以獲得良好的效果,二者修復(fù)效果無顯著差異。
[Abstract]:Background: due to the lack of regeneration ability of chondrocytes, the choice of suitable seed cells is the most important problem in repairing cartilage defects. Aim: to investigate the effect of cartilage formation induced by allogeneic and xenogeneic bone marrow mesenchymal stem cells (MSCs) in repairing laryngeal cartilage defects. Methods: the third generation of human bone marrow mesenchymal stem cells and rabbit bone marrow mesenchymal stem cells were added into the chondrocyte directed inducer (TGF- 尾 1 and bone morphogenetic protein) to induce cartilage formation. It was added to the poly (lactic acid-glycolic acid) copolymer scaffold.
