噴砂酸蝕對鈦鈮鋯錫合金表面成骨細(xì)胞粘附、增殖和分化的影響

【摘要】:目的:研究噴砂酸蝕(SLA)對鈦及鈦鈮鋯錫合金(Ti-24Nb-4Zr-7.9Sn,TNZS)表面形貌的影響,觀察合金的形貌學(xué)特征,評價其生物相容性。方法:將試樣分為鈦機械打磨并拋光組(Ti組),鈦鈮鋯錫機械打磨并拋光組(TNZS組),鈦噴砂酸蝕組(Ti-SLA組)和鈦鈮鋯錫噴砂酸蝕組(TNZS-SLA組),共4組。通過掃描電鏡觀察各組試樣的表面形貌,3D激光共聚焦顯微鏡和接觸角測量儀測量各組試樣表面的粗糙度與親水性。接種MC3T3-E1小鼠前成骨細(xì)胞于各組試樣表面,檢測細(xì)胞在試樣表面的粘附、增殖與礦化的能力,評估其生物相容性。結(jié)果:SLA處理后在材料表面形成納米級及微米級的凹坑,產(chǎn)生均勻分布的粗糙結(jié)構(gòu),經(jīng)過處理后材料仍保持親水性。細(xì)胞在TNZS組上短期粘附明顯高于其它組(P0.05),TNZS-SLA組細(xì)胞增殖、分化能力均明顯高于其它組(P0.05)。結(jié)論:噴砂酸蝕后材料表面相對于光滑材料表面能更有效的促進成骨細(xì)胞在其表面增殖、分化,經(jīng)噴砂酸蝕的鈦鈮鋯錫合金具有良好的細(xì)胞相容性。
[Abstract]:Aim: to investigate the effect of sand blast etching (SLA) on the surface morphology of titanium and Ti-24Nb-4Zr-7.9 SnNZS, to observe the morphologic characteristics of the alloys and to evaluate their biocompatibility. Methods: the samples were divided into four groups: titanium mechanical grinding and polishing group (Ti group), titanium niobium zirconium tin mechanical grinding and polishing group (TNZS group), titanium sandblasting acid etching group (Ti-SLA group) and titanium niobium zirconium tin sandblasting etching group (TNZS-SLA group). The surface morphology of each group was observed by scanning electron microscope (SEM). The surface roughness and hydrophilicity of each group were measured by 3D laser confocal microscope and contact angle measuring instrument. MC3T3-E1 mouse preosteoblasts were inoculated on the surface of each group. The ability of adhesion, proliferation and mineralization of the cells on the surface of the sample was detected, and the biocompatibility of the cells was evaluated. Results the nano-scale and micron size pits were formed on the surface of the material after the treatment of w / SLA, resulting in the uniform distribution of rough structure, and the hydrophilicity of the material was maintained after the treatment. The short-term adhesion of cells in TNZS group was significantly higher than that in other groups (P0.05), and the differentiation ability of cells in TNZS-SLA group was significantly higher than that in other groups (P0.05). Conclusion: compared with smooth surface, the surface of sand-blast acid etched material can promote the proliferation and differentiation of osteoblasts on its surface, and the titanium, niobium, zirconium and tin alloy etched by sandblasting has good cytocompatibility.
