絲素肽結(jié)構(gòu)與止血性能的研究

【摘要】:日常生活中對(duì)突發(fā)性事故的急救治療,醫(yī)院里對(duì)病人手術(shù)過程中的創(chuàng)傷止血,戰(zhàn)爭(zhēng)中對(duì)受傷戰(zhàn)士的現(xiàn)場(chǎng)救護(hù),對(duì)于傷者局部有效的快速止血非常重要。未受控制的出血已經(jīng)成為威脅傷員生命,導(dǎo)致突發(fā)事故、醫(yī)療手術(shù)或戰(zhàn)場(chǎng)死亡的主要原因?焖儆行У闹寡牧峡纱蠓冉档蛡麊T的死亡率。因此,開發(fā)快速、安全、有效的新型止血材料成為醫(yī)學(xué)材料領(lǐng)域的重要研究課題。 絲素蛋白作為從蠶絲中提取的一種天然高分子材料,具有無毒、炎癥反應(yīng)低、生物相容性良好、可生物降解等特點(diǎn),廣泛應(yīng)用于藥物控釋、人造皮膚、人造韌帶以及組織工程等領(lǐng)域。絲素蛋白的水解產(chǎn)物絲素肽,在日用化工、食品加工以及生物醫(yī)藥等領(lǐng)域均有開發(fā)應(yīng)用,但絲素肽在止血材料的研究應(yīng)用尚未見報(bào)道,F(xiàn)階段對(duì)于粉狀止血材料的研究較少,鑒于粉狀止血材料使用及存儲(chǔ)方便,對(duì)其深入的研究探討很有必要。 本文采用CaCl2-乙醇-水(摩爾比1:2:8)三元體系、磷酸和氫氧化鈉對(duì)脫膠蠶絲進(jìn)行水解,通過噴霧干燥和冷凍干燥制備不同分子量的絲素肽,對(duì)其結(jié)構(gòu)和性能進(jìn)行測(cè)試分析,并用于止血實(shí)驗(yàn)和體外細(xì)胞毒性實(shí)驗(yàn),初步研究并探討了絲素肽的理化性質(zhì)和止血性能之間的關(guān)系,主要研究?jī)?nèi)容如下: 1.以CaCl2-乙醇-水(摩爾比1:2:8)三元體系為水解溶液,以58℃,68℃,78℃,88℃,98℃五檔水解溫度和1h,3h,9h,24h四種水解時(shí)間為不同條件水解脫膠蠶絲,通過噴霧干燥和冷凍干燥制備鹽解絲素肽。采用凝膠過濾色譜(GPC)、傅里葉紅外變換光譜(FTIR)、X-射線衍射(XRD)、掃描電鏡(SEM)和氨基酸組分分析等測(cè)試手段對(duì)絲素肽的分子量分布、聚集態(tài)結(jié)構(gòu)、表面形貌及氨基酸組成等進(jìn)行表征。結(jié)果表明,隨著水解溫度的提高和水解時(shí)間的延長(zhǎng),絲素肽的分子量降低,絲素肽的聚集態(tài)結(jié)構(gòu)以無規(guī)卷曲為主,存在少量的β-折疊結(jié)構(gòu),絲素肽粉末形貌以表面褶皺的球囊狀顆粒為主,存在球形顆粒,粒徑隨著分子量的降低變小且更為均勻,主要氨基酸組分包括絲氨酸、甘氨酸、丙氨酸和酪氨酸四種,其組分及含量與水解溫度和水解時(shí)間無明顯關(guān)聯(lián); 2.以磷酸溶液為水解溶液,設(shè)計(jì)磷酸濃度、水解溫度和水解時(shí)間三因素正交水解實(shí)驗(yàn),通過冷凍干燥制備酸解絲素肽粉末。采用同樣的測(cè)試手段對(duì)酸解絲素肽進(jìn)行性能表征。結(jié)果表明,酸解絲素肽的分子量明顯降低,主要分布在3.8~12.6kDa,絲素肽的聚集態(tài)結(jié)構(gòu)為無規(guī)卷曲,絲素肽粉末呈現(xiàn)無規(guī)碎片狀,主要氨基酸組分包括甘氨酸、丙氨酸、天冬氨酸和谷氨酸等。 3.以氫氧化鈉溶液為水解溶液,設(shè)計(jì)氫氧化鈉濃度、水解溫度和水解時(shí)間三因素正交水解實(shí)驗(yàn),通過冷凍干燥制備絲素肽粉末。測(cè)試手段同上所述。結(jié)果表明,堿解絲素肽的分子量較低,分布集中在3.6~18.9kDa,絲素肽的聚集態(tài)結(jié)構(gòu)為無規(guī)卷曲,絲素肽粉末表面形貌與酸解絲素肽相似,,主要氨基酸組分為天冬氨酸、谷氨酸、丙氨酸和酪氨酸等。 4.不同水解方法得到的絲素肽的止血性能及體外細(xì)胞毒性研究。通過凝血彈性描記(TEG)測(cè)試、大鼠肝臟創(chuàng)傷止血模型和體外細(xì)胞毒性實(shí)驗(yàn)表征絲素肽的止血性能和生物相容性。結(jié)果表明,高分子量鹽解絲素肽具備與Arista相近的止血性能,而低分子量鹽解絲素肽、酸解絲素肽和堿解絲素肽則無顯著的止血性能;各種水解方法制備的絲素肽對(duì)細(xì)胞的生長(zhǎng)繁殖無毒副作用。
[Abstract]:The rapid , safe and effective hemostatic material is an important research topic in the field of medical materials .

Silk fibroin is a kind of natural high molecular material extracted from silk , and has the characteristics of no toxicity , low inflammation , good biocompatibility , biodegradability and the like , and is widely applied to the fields of drug controlled release , artificial skin , artificial ligament and tissue engineering .

In this paper , CaCl2 - ethanol - water ( molar ratio 1 : 2 : 8 ) ternary system , phosphoric acid and sodium hydroxide were used to hydrolyze the degumming silk . The structure and properties of silk fibroin peptides with different molecular weights were prepared by spray drying and freeze drying . The relationship between the physicochemical properties and hemostatic properties of silk fibroin peptide was studied . The main contents of the study were as follows :

The molecular weight distribution , aggregation state structure , surface morphology and amino acid composition of silk fibroin peptide were characterized by means of spray drying and freeze drying .


2 . The acid solution was prepared by orthogonal hydrolysis with phosphoric acid solution as hydrolysis solution , phosphoric acid concentration , hydrolysis temperature and hydrolysis time . The results showed that the molecular weight of the fibroin peptide was obviously reduced , the main distribution was 3.8 - 12.6 kDa , the aggregation state of silk fibroin peptide was random , and the main amino acid component included glycine , alanine , aspartic acid and glutamic acid .

3 . Using sodium hydroxide solution as hydrolysis solution , designing sodium hydroxide concentration , hydrolysis temperature and hydrolysis time three factors orthogonal hydrolysis experiment , and preparing silk fibroin powder by freeze - drying . The results showed that the molecular weight of silk fibroin peptide was lower , the distribution concentration was 3.6 - 18.9 kDa , the aggregation state of silk fibroin peptide was 3 . 6 - 18 . 9 kDa , the morphology of silk fibroin peptide was similar to that of acid - releasing silk peptide , and the main amino acid component was aspartic acid , glutamic acid , alanine and tyrosine .

4 . The hemostatic properties and in vitro cytotoxicity of silk fibroin peptides obtained by different methods of hydrolysis were studied . The hemostatic properties and biocompatibility of silk fibroin peptides were characterized by thromboelastography ( TEG ) test , rat liver trauma hemostasis model and in vitro cytotoxicity experiment .
The silk fibroin peptide prepared by various hydrolysis methods has no toxic or side effect on the growth and propagation of cells .
