分層多功能層層自組裝肝素表面涂層的制備和血液相容性檢測

【摘要】:背景:心室輔助裝置在臨床應(yīng)用取得了良好的治療效果,同時也面臨著長期使用引起的出血、感染、血栓形成等并發(fā)癥。要減少這些并發(fā)癥,都亟待改進(jìn)心室輔助裝置與血液接觸面的血液相容性。涂層技術(shù)可以在不改變基材的前提下改善材料表面性質(zhì),提高血液相容性,從而減少人工植入裝置植入后的不良反應(yīng)。 目的:制備分層多功能層層自組裝肝素涂層,并對照研究該涂層的血液相容性及抗凝特性與傳統(tǒng)涂層的差別。 方法:利用層層自組裝技術(shù)組裝出有底層和表層兩個功能層的多層自組裝涂層,底層為殼聚糖層,通過共價鍵固定在鈦金屬表面,表層為多層肝素/Fe3+層層自組裝涂層,共5層肝素。以組裝相同層數(shù)的肝素/殼聚糖層層自組裝涂層作為多層對照組;單層對照組為肝素和殼聚糖共混溶液的單層涂層;空白對照組為裸鈦片。對照研究不同涂層的肝素含量、肝素釋放量,以及血液相容性的差別。 結(jié)果:測定分層多功能自組裝涂層的肝素總含量為:7.57±0.55μg/cm2,優(yōu)于多層對照組和單層對照組的5.13±0.57μg/cm2和1.55±0.79μg/cm2(P0.05),同時實(shí)驗(yàn)組4,12,24,48小時肝素溶解釋放量均明顯高于多層對照組和單層對照組。實(shí)驗(yàn)制備的分層多功能涂層在溶血率測定也優(yōu)于多層對照組和空白對照組,同時5%的溶血率符合國家標(biāo)準(zhǔn)。動態(tài)凝血實(shí)驗(yàn)顯示實(shí)驗(yàn)制備的分層多功能自組裝涂層抗凝性能最好,每個時間點(diǎn)抗凝血程度均優(yōu)于對照組。 結(jié)論:分層多功能層層自組裝肝素涂層在相同的組裝條件和組裝層數(shù)時,能夠組裝更多的肝素,同時有利于肝素的溶解釋放,能夠提高材料的血液相容性和抗凝血性能。
[Abstract]:Background: ventricular assistive device has achieved good therapeutic effect in clinical application, and also faces complications such as bleeding, infection and thrombosis caused by long-term use. To reduce these complications, it is necessary to improve the blood compatibility of ventricular assistive devices and blood interface. The coating technology can improve the surface properties of the material without changing the substrate and improve the blood compatibility, thus reducing the adverse reactions after the implantation of the artificial implants. Aim: to prepare layered multifunctional layer self assembled heparin coating and compare the blood compatibility and anticoagulant characteristics between the coating and the traditional coating. Methods: the multilayer self-assembly coating with two functional layers was assembled by layer self-assembly technique. The bottom layer was chitosan layer, which was fixed on titanium surface by covalent bond, and the surface layer was multi-layer heparin / Fe _ 3 layer self-assembly coating. A total of 5 layers of heparin. The layers of heparin / chitosan self-assembled coatings with the same number of layers were used as the multilayer control group; the monolayer control group was the monolayer coating of heparin and chitosan blend solution; and the blank control group was a bare titanium sheet. The heparin content, heparin release and blood compatibility of different coatings were studied. Results: the total heparin content of the multifunctional self-assembled coating was 7. 57 鹵0. 55 渭 g / cm ~ 2, which was better than that of the multilayer control group and the monolayer control group (5. 13 鹵0. 57 渭 g/cm2 and 1. 55 鹵0. 79 渭 g / cm ~ (2) P 0. 05). Meanwhile, the heparin release of the experimental group at 48 hours was significantly higher than that of the multilayer control group and the monolayer control group. The multifunctional coating was also superior to the multilayer control group and the blank control group in the determination of hemolysis rate, and the hemolysis rate of 5% was in line with the national standard. The dynamic coagulation experiment showed that the layered multifunctional self-assembled coating had the best anticoagulant performance and the anticoagulant degree at each time point was better than that of the control group. Conclusion: the layered multifunctional layer self-assembled heparin coating can assemble more heparin at the same assembly conditions and the number of the assembled layers, and it is beneficial to the dissolution and release of heparin, and can improve the blood compatibility and anticoagulant performance of the materials.
