當(dāng)歸多糖膠原緩釋微球復(fù)合新型生物活性多孔支架的制備及其細(xì)胞相容性的研究

【摘要】:目的制備并研究當(dāng)歸多糖明膠緩釋微球復(fù)合硫酸鈣/凍干骨支架的細(xì)胞相容性。方法支架用煅燒掛漿法制備,分別在原支架和表面修飾后的支架上復(fù)合培養(yǎng)第三代成骨細(xì)胞,利用倒置顯微鏡觀察兩種支架上的成骨細(xì)胞數(shù)目及形態(tài)等,利用掃描電鏡觀察復(fù)合支架和成骨細(xì)胞復(fù)合支架培養(yǎng)時(shí)的支架表面;評(píng)估方法為細(xì)胞蛋白質(zhì)和細(xì)胞黏附率測(cè)定法。結(jié)果在復(fù)合成骨細(xì)胞培養(yǎng)的各個(gè)時(shí)期,實(shí)驗(yàn)組的蛋白質(zhì)含量較對(duì)照組含量多,并且實(shí)驗(yàn)組的細(xì)胞黏附率也高于對(duì)照組(P0.05)。提示新型支架較原支架更有利于細(xì)胞生長(zhǎng),并且電鏡下觀察到支架孔隙中也有細(xì)胞生長(zhǎng)。結(jié)論經(jīng)過(guò)當(dāng)歸多糖修飾后的支架較原支架的細(xì)胞相容性高。
[Abstract]:Objective to prepare and study the cytocompatibility of Angelica polysaccharide gelatin sustained-release microspheres combined with calcium sulfate / freeze-dried bone scaffold. Methods the scaffolds were prepared by calcination-suspension method. The third generation osteoblasts were co-cultured on the original scaffold and the surface modified scaffold respectively. The number and morphology of osteoblasts on the scaffolds were observed by inverted microscope. The surface of composite scaffold and osteoblast composite scaffold was observed by scanning electron microscope (SEM). Results the protein content of the experimental group was higher than that of the control group, and the cell adhesion rate of the experimental group was higher than that of the control group. The results indicated that the new scaffold was more favorable to cell growth than the original scaffold, and the cell growth was observed in the pore of the scaffold under electron microscope. Conclusion the scaffold modified by Angelica polysaccharide has higher cytocompatibility than the original scaffold.
