含銅不銹鋼銅離子釋放機(jī)制研究及其生物學(xué)效應(yīng)探索

【摘要】:在醫(yī)用金屬材料中,醫(yī)用不銹鋼具有優(yōu)良的耐腐蝕性能、高強韌性、易加工和成本低等優(yōu)點,成為目前臨床上應(yīng)用最廣泛醫(yī)用金屬材料之一。然而,目前的醫(yī)用不銹鋼不具有生物活性。外科植入手術(shù)中細(xì)菌感染是導(dǎo)致手術(shù)失敗的主要原因。隨著人們對生活質(zhì)量的要求越來越高,醫(yī)用金屬材料需要具有良好的抗菌性能。銅元素不但是人體必須的微量元素,還具有良好的殺菌性能。在普通不銹鋼中添加適量的銅,通過熱處理的方式使含銅不銹鋼在使用時釋放銅離子,使含銅不銹鋼具有良好的生物功能和抗菌性能。本文對含銅不銹鋼的銅離子釋放機(jī)制和其生物功能進(jìn)行了探索,得到了以下研究成果: (1)含銅不銹鋼的銅離子溶出機(jī)制 含銅不銹鋼的表面鈍化膜與普通不銹鋼不同,銅離子參與鈍化膜的形成,并隨鈍化膜的腐蝕而溶出。由于含銅不銹鋼中析出富銅相,含銅不銹鋼的耐點蝕性能略低于普通不銹鋼,但是耐均勻腐蝕性與普通不銹鋼相近。經(jīng)過硝酸鈍化后含銅不銹鋼的耐點蝕性能與普通不銹鋼相差無幾。 (2)含銅不銹鋼的抗菌性能 含銅不銹鋼對生活中和外科植入手術(shù)造成感染的常見細(xì)菌:金黃色葡萄球菌和大腸桿菌,具有良好的殺滅作用。含銅不銹鋼對生物膜的形成也具有良好的抑制作用。硝酸鈍化后含銅不銹鋼的殺菌性能有所下降。通過用硫酸銅溶液代替不銹鋼溶出的銅離子,得到銅離子的最低殺菌濃度(MBC)為50ppb。 (4)含銅不銹鋼的生物相容性 含銅不銹鋼的溶血率小于5%,不會發(fā)生溶血,表明銅元素的添加不會對不銹鋼的血液相容性造成影響。生物安全性評價說明317L-4.5Cu不銹鋼沒有細(xì)胞毒性,符合植入標(biāo)準(zhǔn)。
[Abstract]:Among the medical metal materials, medical stainless steel has the advantages of excellent corrosion resistance, high strength and toughness, easy processing and low cost, so it has become one of the most widely used medical metal materials in clinic. However, the current medical stainless steel does not have biological activity. Bacterial infection in surgical implantation is the main cause of failure. With the increasing demand for quality of life, medical metal materials need to have good antibacterial properties. Copper is not only a necessary trace element for human body, but also has good bactericidal performance. By adding proper amount of copper to common stainless steel, copper ion is released from copper containing stainless steel by heat treatment, which makes copper stainless steel have good biological function and antibacterial property. In this paper, the mechanism of copper ion release and its biological function of copper containing stainless steel have been explored, and the following research results have been obtained: Copper ion dissolution mechanism of copper containing stainless steel The surface passivation film of copper-containing stainless steel is different from that of common stainless steel. Copper ion takes part in the formation of passivation film and dissolves with the corrosion of passivation film. Due to the copper-rich phase in the copper-containing stainless steel, the pitting corrosion resistance of the copper-containing stainless steel is slightly lower than that of the common stainless steel, but the uniform corrosion resistance is similar to that of the common stainless steel. After passivation with nitric acid, the pitting corrosion resistance of copper containing stainless steel is similar to that of ordinary stainless steel. Antibacterial properties of copper containing stainless steel Copper-containing stainless steel has a good killing effect on common bacteria in life and surgical implantation: Staphylococcus aureus and Escherichia coli. Copper-containing stainless steel also has a good inhibitory effect on the formation of biofilm. The bactericidal properties of copper-containing stainless steel decreased after passivation with nitric acid. The minimum bactericidal concentration of copper ion (MBC) is 50 ppb by replacing the copper ion dissolved in stainless steel with copper sulfate solution. Biocompatibility of copper containing stainless steel The hemolysis rate of the stainless steel containing copper is less than 5 and the hemolysis will not occur, which indicates that the addition of copper does not affect the blood compatibility of the stainless steel. Biosafety evaluation shows that 317L-4.5Cu stainless steel is not cytotoxic and meets the implantation criteria.
