干細(xì)胞3D支架的研究進(jìn)展

【摘要】:干細(xì)胞是一類具有無限增殖潛能,可自我更新的細(xì)胞,不同的培養(yǎng)或誘導(dǎo)條件下能夠分化成為不同的細(xì)胞類型。目前,隨著醫(yī)學(xué)治療手段及干細(xì)胞研究的不斷發(fā)展,發(fā)現(xiàn)干細(xì)胞可應(yīng)用于很多疾病的治療,干細(xì)胞臨床應(yīng)用日益廣泛,逐漸成為科研工作者研究的熱點(diǎn)。然而干細(xì)胞移植進(jìn)入生物體后繁殖效率很低,無法達(dá)到需求的量,因此如何對(duì)干細(xì)胞在生物體外進(jìn)行大量培養(yǎng)擴(kuò)增、在生物體內(nèi)如何更穩(wěn)定地增殖分化成為迫切需要解決的難題。當(dāng)前干細(xì)胞最主要的培養(yǎng)方法仍是2D培養(yǎng),2D培養(yǎng)無法模擬體內(nèi)的3D微環(huán)境,繁殖效率較低,正是由于2D培養(yǎng)局限性太多,促使國(guó)內(nèi)外學(xué)者對(duì)3D培養(yǎng)技術(shù)和3D支架材料進(jìn)行深入探索,并取得了大量成果。對(duì)3D培養(yǎng)技術(shù)在干細(xì)胞中的發(fā)展與應(yīng)用進(jìn)行概述和展望。
[Abstract]:Stem cells are a kind of cells with unlimited proliferative potential and self-renewal. They can differentiate into different cell types under different culture or induction conditions. At present, with the development of medical treatment and stem cell research, it is found that stem cells can be used in the treatment of many diseases. However, stem cell transplantation into the organism after reproduction efficiency is very low, can not reach the required amount, so how to expand the stem cells in vitro culture in a large number of organisms, How to more steadily proliferate and differentiate in organisms has become an urgent problem to be solved. At present, the main method of stem cell culture is that 2D culture can't simulate 3D microenvironment in vivo, and its reproductive efficiency is low, because 2D culture has too many limitations. In order to promote the domestic and foreign scholars to explore the 3D culture technology and 3D scaffold materials, and achieved a lot of results. The development and application of 3D culture technology in stem cells were reviewed and prospected.
