【摘要】:目的研究三維鈦網(wǎng)支架(TW)材料對(duì)犬牙周膜成纖維附著、增殖、分化的影響。方法通過(guò)酶消化組織塊法原代培養(yǎng)犬牙周膜成纖維細(xì)胞(PDLFs),傳代后與TW材料接觸培養(yǎng),噻唑藍(lán)(MTT)法比較實(shí)驗(yàn)組三維鈦網(wǎng)培養(yǎng)與對(duì)照組二維培養(yǎng)對(duì)種子細(xì)胞的促增殖作用,通過(guò)提取測(cè)量上清液堿性磷酸酶含量比較實(shí)驗(yàn)組三維鈦網(wǎng)培養(yǎng)與對(duì)照組傳統(tǒng)礦化誘導(dǎo)液培養(yǎng)對(duì)種子細(xì)胞的促分化作用。結(jié)果酶消化組織塊法可以成功獲得用于實(shí)驗(yàn)的犬PDLFs,且細(xì)胞能有效的與TW附著,實(shí)驗(yàn)組的促增殖和分化作用要顯著優(yōu)于對(duì)照組。結(jié)論 TW材料能有效促進(jìn)牙PDLFs的附著、增殖、分化,在牙周組織再生工程有良好的應(yīng)用前景。
[Abstract]:Objective to study the effect of three dimensional titanium mesh scaffold (TW) on the attachment, proliferation and differentiation of canine periodontic membrane.Methods using enzyme digestion tissue block method, the primary cultured fibroblasts of perodontoid membrane were cultured in contact with the TW material. The proliferation of seed cells in the experimental group was compared with that in the control group by the method of MTT- (thiazolyl blue), three dimensional titanium mesh culture and the control group.By extracting and measuring the content of alkaline phosphatase in supernatant, the effects of three dimensional titanium mesh culture on the differentiation of seed cells in the experimental group and the traditional mineralized inducer culture in the control group were compared.Results PDLFs could be successfully obtained by enzyme digestion tissue block method, and the cells could attach to TW effectively. The effect of promoting proliferation and differentiation in the experimental group was significantly better than that in the control group.Conclusion TW can effectively promote the attachment, proliferation and differentiation of tooth PDLFs, and has a good application prospect in periodontal tissue regeneration engineering.
