不同復(fù)溫方法對(duì)深低溫冷凍復(fù)合組織血管活性的影響

【摘要】:目的比較3種不同復(fù)溫方法對(duì)深低溫冷凍的復(fù)合組織中血管活性的影響,找出一種對(duì)復(fù)合組織中血管損傷最小的復(fù)溫方法。方法選用20只新西蘭大白兔隨機(jī)分成四組,切取雙側(cè)后肢(對(duì)照組為新鮮斷肢取血管),三個(gè)實(shí)驗(yàn)組標(biāo)本經(jīng)梯度降溫后放入液氮中保存7天。分別采用37℃水浴復(fù)溫組(A組)、空氣復(fù)溫組(B組)、-20℃冰復(fù)溫組(C組)方法進(jìn)行復(fù)溫。采用光鏡、免疫組化方法觀察血管內(nèi)皮細(xì)胞活性,結(jié)果進(jìn)行統(tǒng)計(jì)學(xué)分析。結(jié)果光鏡觀察,A組、B組內(nèi)皮細(xì)胞大片脫落,平滑肌細(xì)胞變形;而C組內(nèi)皮細(xì)胞和平滑肌細(xì)胞保存較好,更接近對(duì)照組;免疫組化檢測(cè)中,3種復(fù)溫方法血管內(nèi)皮中均可見VEGF及CD34呈斑片狀的弱表達(dá),-20℃冰復(fù)溫組(C組)VEGF及CD34的表達(dá)更接近對(duì)照組,高于37℃水浴復(fù)溫組(A組)及空氣復(fù)溫組(B組)(P0.05)。結(jié)論 3種不同復(fù)溫方法對(duì)深低溫冷凍的復(fù)合組織中血管活性影響不同,-20℃冰復(fù)溫法影響最小。
[Abstract]:Objective to compare the effects of three different rewarming methods on vascular activity in deep hypothermia and cryopreservation composite tissues, and to find out a rewarming method with minimal vascular damage in the composite tissues.
