超支化聚合物新材料富集糖肽方法的研究

【摘要】:目的:將自合成的超支化聚合物固定到氨基硅球上形成一種新型的糖肽富集材料,用于糖肽的高效分離與富集。方法:陰離子開環(huán)聚合法合成的超支化聚縮水甘油,經高碘酸氧化成為超支化聚甘油醛(hyperbranched polyglycerol-aldehydes,HPG-ALD)后固定于氨基硅球上,再用己二酸二酰肼修飾得到的酰肼材料應用于糖肽富集。結果:經掃描電鏡(Scanning Electron Microscope,SEM)、熱重分析(thermogravimetric analysis,TGA)等手段證明材料被成功合成。同時,使用制備成功的新材料來富集標準糖蛋白和糖肽都獲得了良好的富集效果。結論:實驗證明超支化縮水甘油酰肼材料(hyperpolymer-assisted hydrazide functionalized microspheres,HHMs)可用于低豐度糖蛋白/糖肽的富集。
[Abstract]:Aim: to immobilize the hyperbranched polymer onto an amino silica sphere to form a novel glycopeptide enrichment material for the efficient separation and enrichment of glycopeptide. Methods: hyperbranched polyglycidyl was synthesized by anion ring-opening polymerization. Hyperbranched polyglycerol-aldehydesy (HPG-ALD) was oxidized by periodate to form hyperbranched polyglycerol-aldehydes.Then the hyperbranched polyglycerol-aldehydes-HPG-ALD was immobilized on the silica spheres. The glycopeptide was enriched by acyl hydrazide modified with dihydrazide adipate. Results: the material was successfully synthesized by scanning electron microscope (SEM), thermogravimetric analysis (TGA), scanning electron microscopy (SEM) and thermogravimetric analysis (TGA). The results show that hyperbranched glycidyl hydrazide (HHMs) can be used in the enrichment of low abundance glycoprotein / glycopeptide. Conclusion: hyperbranched glycidyl hydrazide (HHMs) can be used in the enrichment of low abundance glycoprotein / glycopeptide.
