【摘要】：背景：骨髓間充質(zhì)干細(xì)胞具有自我復(fù)制和多向分化的潛能,我們已利用骨髓間充質(zhì)干細(xì)胞成功誘導(dǎo)出血管內(nèi)皮細(xì)胞和血管平滑肌細(xì)胞,并證實(shí)所誘導(dǎo)的平滑肌細(xì)胞和膠原包埋的聚乙醇酸支架具有良好的組織相容性。目的：利用骨髓間充質(zhì)干細(xì)胞誘導(dǎo)血管壁種子細(xì)胞,體外構(gòu)建初級組織工程化血管模型,植入動物皮下探討利用動物皮下作為生物反應(yīng)器構(gòu)建小口徑組織工程化血管的可行性。方法：骨髓間充質(zhì)干細(xì)胞誘導(dǎo)為血管平滑肌樣細(xì)胞和血管內(nèi)皮樣細(xì)胞,分層種植于膠原包埋聚乙醇酸支架表面,將細(xì)胞-支架復(fù)合體種植于動物皮下,于植入后第4、8周取出植入皮下的組織工程化小口徑血管,行組織學(xué)檢查及免疫熒光檢查。結(jié)果：組織學(xué)分析顯示組織工程化小口徑血管壁一個典型的動脈結(jié)構(gòu),包括內(nèi)皮組織、中膜和外膜。8周后,植入的種子細(xì)胞標(biāo)記Brdu會發(fā)現(xiàn)血管壁三層。結(jié)論：皮下組織構(gòu)建組織工程化小口徑血管是一個很好的生物反應(yīng)器。組織工程血管的解剖類似于自然的血管。
[Abstract]:Background: bone marrow mesenchymal stem cells have the potential of self-replication and multi-differentiation. We have successfully induced vascular endothelial cells and vascular smooth muscle cells using bone marrow mesenchymal stem cells. It was confirmed that the induced smooth muscle cells and collagen-coated polyglycolic acid scaffolds had good histocompatibility. Objective: to induce vascular wall seed cells from bone marrow mesenchymal stem cells (BMSCs) and to construct primary tissue engineered vascular models in vitro. To explore the feasibility of using animal subcutaneous as bioreactor to construct small caliber tissue engineered blood vessels methods: bone marrow mesenchymal stem cells were induced into vascular smooth muscle cells and vascular endothelioid cells. The cell-scaffold complex was implanted subcutaneously on the surface of collagen-coated polyglycolic acid scaffold, and the tissue engineering vessels were removed 8 weeks after implantation. Results: histological analysis showed that a typical arterial structure of tissue engineered small diameter vascular wall, including endothelial tissue, medial and adventitia, was observed at 8 weeks later. Three layers of vascular wall could be found by seeded cells labeled Brdu. Conclusion: subcutaneous tissue construction of small tissue engineering vessels is a good bioreactor. The anatomy of tissue engineered blood vessels is similar to that of natural vessels.
