骨生物材料復(fù)合骨髓間充質(zhì)干細(xì)胞異位成骨修復(fù)肋骨大段缺損

【摘要】:背景:組織工程生物材料具有與自體組織相似的結(jié)構(gòu)與功能。目的:觀察骨生物材料復(fù)合大鼠骨髓間充質(zhì)干細(xì)胞異位成骨治療肋骨大段缺損的效果。方法:取Wistar大鼠50只,制備右側(cè)大段肋骨缺損模型,隨機(jī)分為2組,對(duì)照組于骨缺損處置入氯化鈣-藻酸鈉凝膠,實(shí)驗(yàn)組于骨缺損處植入氯化鈣-藻酸鈉-骨髓間充質(zhì)干細(xì)胞復(fù)合物。植入后2,4,8周,進(jìn)行胸部X射線及骨缺損處組織形態(tài)學(xué)觀察。結(jié)果與結(jié)論:1X射線觀察結(jié)果:植入后2周,實(shí)驗(yàn)組骨缺損區(qū)無明顯變化;植入后4周,缺損部位開始出現(xiàn)骨質(zhì)痕跡;植入后8周,缺損部位大部分充滿骨質(zhì),缺損兩端逐漸相接。對(duì)照組缺損部位始終未見明顯骨質(zhì)修復(fù),缺損兩端逐漸封閉并硬化;2骨缺損處組織形態(tài)學(xué)觀察結(jié)果:植入后2周,實(shí)驗(yàn)組材料腔隙中可見少量骨纖維組織,大量炎性細(xì)胞聚集,材料與骨質(zhì)末端無連接;對(duì)照組骨缺部位可見大量炎性細(xì)胞,未見骨組織形成。植入后4周,實(shí)驗(yàn)組支架逐漸降解,可見大量新生骨質(zhì);對(duì)照組支架僅少量降解,缺損兩端出現(xiàn)骨組織聚集。植入后8周,實(shí)驗(yàn)組支架大部分降解,可見大量骨組織、骨小梁等組織結(jié)構(gòu),缺損兩端與再生骨質(zhì)連接;對(duì)照組支架大部分降解,缺損兩端出現(xiàn)骨硬化;3結(jié)果表明:骨生物材料復(fù)合大鼠骨髓間充質(zhì)干細(xì)胞可促進(jìn)肋骨大段缺損的修復(fù)。
[Abstract]:Background: tissue engineering biomaterials have the same structure and function as autologous tissues. Objective: to observe the effect of bone biomaterials combined with bone marrow mesenchymal stem cells (BMSCs) on heterotopic osteogenesis of rib defects. 50 Wistar rats were selected. The right large rib defect model was made and divided into two groups randomly. The control group was treated with calcium chloride sodium alginate gel in the treatment of bone defect. The experimental group was implanted with calcium chloride-sodium alginate-bone marrow mesenchymal stem cell complex at the site of bone defect. Results and conclusion the results showed that there was no significant change in bone defect area in the experimental group 2 weeks after implantation. 4 weeks after implantation, bone marks began to appear in the defect. At 8 weeks after implantation, most of the defects were filled with bone, and the two ends of the defects were gradually connected with each other. In the control group, there was no obvious bone repair at the defect site, and the two ends of the defect were gradually closed and sclerotic. 2Histomorphologic observation of bone defect: at 2 weeks after implantation, a small amount of bone fiber tissue was found in the space of the experimental group, and a large number of inflammatory cells gathered, and there was no connection between the material and the end of the bone. In the control group, a large number of inflammatory cells were found in the bone defect, but no bone tissue was formed. 4 weeks after implantation, the scaffolds in the experimental group were gradually degraded and a large number of new bone was observed. In the control group, there was only a small amount of degradation and bone tissue aggregation at the two ends of the defect. 8 weeks after implantation, most of the scaffolds in the experimental group were degraded, a large number of bone tissues and trabeculae were found, and the two ends of the defect were connected with the regenerated bone. In the control group, most of the scaffolds degraded and bone sclerosis appeared at both ends of the defect. The results showed that bone biomaterials combined with bone marrow mesenchymal stem cells could promote the repair of rib defect.
