周期性拉伸應(yīng)力與人永生化角質(zhì)形成細(xì)胞的黏附和鋪展

【摘要】:背景:皮膚組織所處的力學(xué)環(huán)境以及上皮細(xì)胞的鋪展?fàn)顟B(tài)與傷口愈合和瘢痕形成過(guò)程關(guān)系密切。目的:分析胞外力學(xué)刺激對(duì)細(xì)胞鋪展的作用,并檢測(cè)細(xì)胞的增殖狀態(tài)進(jìn)一步分析鋪展形態(tài)對(duì)細(xì)胞增殖等生理活動(dòng)的影響。方法:通過(guò)FX-4000柔性基底加載系統(tǒng)對(duì)人永生化角質(zhì)形成細(xì)胞(HaCaT)進(jìn)行周期性拉伸應(yīng)力正弦波加載,加載程式0.2 Hz,10%拉伸幅度。在0,24,48 h對(duì)細(xì)胞的鋪展形態(tài)進(jìn)行對(duì)比,利用流式細(xì)胞儀檢測(cè)分析細(xì)胞的增殖,并利用免疫熒光染色方法對(duì)比分析紐蛋白在細(xì)胞內(nèi)的分布。結(jié)果與結(jié)論:HaCaT細(xì)胞在加載24 h后分裂期細(xì)胞較多,鋪展形態(tài)無(wú)明顯變化;加載48 h后HaCaT細(xì)胞形態(tài)發(fā)生較為明顯的變化,分裂期細(xì)胞較靜態(tài)對(duì)照組略有減少;拉伸應(yīng)力作用下紐蛋白的分布由細(xì)胞核周圍的膜區(qū)域向細(xì)胞邊緣集中。結(jié)果表明:適度的力學(xué)加載能夠在保持細(xì)胞鋪展和黏附狀態(tài)下,促進(jìn)細(xì)胞增殖;周期性持續(xù)拉伸應(yīng)力的作用時(shí)間是HaCaT細(xì)胞鋪展形態(tài)和與基底黏附位點(diǎn)的重要影響因素。
[Abstract]:Background: the mechanical environment of skin tissue and the spreading state of epithelial cells are closely related to wound healing and scar formation. Objective: to analyze the effect of extracellular mechanical stimulation on cell spreading. The effects of spreading morphology on cell proliferation and other physiological activities were analyzed. Methods: human immortalized keratinocytes were treated by FX-4000 flexible substrate loading system. HaCaT is subjected to sine wave loading of cyclic tensile stress. The extension range of 0.2 Hzn was 10%. The proliferation of the cells was analyzed by flow cytometry after 48 hours. The distribution of neutropin in the cells was analyzed by immunofluorescence staining. Results and conclusion: after 24 hours of loading, there were more cells in the mitotic phase, but no obvious changes in the spreading morphology of the cells. After 48 h of loading, the morphologic changes of HaCaT cells were more obvious, and the number of cells in the mitotic phase was slightly lower than that in the static control group. Under tensile stress, the distribution of Neuron was concentrated from the membrane area around the nucleus to the edge of the cell. The results showed that moderate mechanical loading could promote cell proliferation while keeping the state of cell spreading and adhesion. The duration of cyclic continuous tensile stress is an important factor affecting the spreading morphology of HaCaT cells and the adhesion sites to the substrate.
