咪喹莫特誘導(dǎo)C3H/HeJ小鼠斑禿動(dòng)物模型的建立及發(fā)病機(jī)理初步研究

目的:探討應(yīng)用咪喹莫特建立C3H/HeJ小鼠斑禿動(dòng)物模型的可行性,對(duì)該方法誘導(dǎo)的斑禿模型與人類斑禿的相似性進(jìn)行評(píng)價(jià),并初步研究咪喹莫特誘導(dǎo)C3H/HeJ小鼠斑禿形成的機(jī)理。方法:76只健康C3H/HeJ小鼠隨機(jī)分成11組。實(shí)驗(yàn)5組,分別給予不同部位和時(shí)間間隔的咪喹莫特乳膏外用;對(duì)照6組,分別給予空白賦形劑外用、不同濃度的IFN-γ靜脈或皮下給藥、不做任何處理,一定時(shí)間后觀察各方法對(duì)斑禿的誘導(dǎo)作用。在皮損特點(diǎn)、臨床經(jīng)過(guò)和組織學(xué)特征等方面對(duì)咪喹莫特誘導(dǎo)的C3H/HeJ小鼠斑禿模型與人類斑禿的相似性進(jìn)行評(píng)估,并取各組小鼠皮膚進(jìn)行抗CD4+T、CD8+T細(xì)胞免疫組化染色和IFN-γmRNA原位雜交實(shí)驗(yàn),以探討咪喹莫特誘導(dǎo)C3H/HeJ小鼠斑禿形成的機(jī)理。結(jié)果:咪喹莫特外用可良好地誘導(dǎo)C3H/HeJ小鼠斑禿模型的建立,尤以每周三次項(xiàng)部的給藥方式為最佳,其成功率和誘導(dǎo)期均優(yōu)于其它各組。由咪喹莫特誘導(dǎo)的斑禿模型在皮損特點(diǎn)、臨床經(jīng)過(guò)和組織學(xué)特征等方面具有與人類斑禿基本相似的臨床特征,給藥后皮膚內(nèi)有CD4+T、CD8+
