BIGLYCAN在光老化中的作用機(jī)制研究

研究背景:二聚糖(Biglycan, BGN)是一種屬于糖蛋白類的蛋白質(zhì),由42kDa的核心蛋白(core protein)和兩個(gè)硫酸軟骨素/硫酸皮膚素(chondroitin sulfate, CS/dermatan sulfate, DS)糖胺聚糖(glycosaminoglycan, GAG)鏈組成,在皮膚組織結(jié)構(gòu)合成、空間填充和皮膚生理功能中起到重要的作用。雖然許多組織表達(dá)豐富的二聚糖,但其生物功能仍然知之甚少。二聚糖核心蛋白的合成起始于內(nèi)質(zhì)網(wǎng)(endoplasmic reticulum, ER),并繼續(xù)轉(zhuǎn)移至高爾基體,通過各種糖基轉(zhuǎn)移酶(glycosyltransferases, GTs)的作用下合成CS/DS-GAG鏈。二聚糖具有組建細(xì)胞外基質(zhì)(extracellular matrix, ECM)的作用,靶向敲除二聚糖基因可以觀察到異常膠原纖維形態(tài)和骨質(zhì)疏松癥樣表型。分泌的二聚糖通過其核心蛋白和GAG側(cè)鏈的共同作用下參與合成ECM,特別是對(duì)Ⅰ、 Ⅱ、Ⅲ、Ⅵ型膠原蛋白和彈力蛋白的裝配起到重要的作用。最近研究發(fā)現(xiàn)二聚糖與炎癥密切相關(guān),在脂多糖(lipopolysaccharid...
