慢性紫外線損傷小鼠模型皮膚角質(zhì)形成細(xì)胞中CK1和CK10的表達(dá)研究

【摘要】:目的:觀察慢性紫外線(UV)損傷對小鼠角質(zhì)形成細(xì)胞(KC)角蛋白(CK)1和CK10表達(dá)的影響。方法:將實(shí)驗(yàn)小鼠隨機(jī)分為空白對照組(20只)和UV損傷組(20只),采用UV光療儀模擬日光中的長波紫外線(UVA)和中波紫外線(UVB)照射實(shí)驗(yàn)小鼠。照射從最小紅斑量起始,UVB為0.07 J/cm~2,UVA為0.7 J/cm~2;每日1次,每周5 d,每周增加0.5個紅斑量,連續(xù)照射9周,UVB累計照射劑量達(dá)9.45 J/cm~2,UVA累計照射劑量達(dá)94.5 J/cm~2。通過蘇木精-伊紅(HE)染色和間苯二酚-堿性品紅染色(Weigert染色法)確定皮膚損傷,通過免疫組化方法分別對照射前(W0)、后(W9)CK1和CK10的表達(dá)進(jìn)行檢測。應(yīng)用配對t檢驗(yàn)和獨(dú)立樣本t檢驗(yàn)對相關(guān)數(shù)據(jù)進(jìn)行統(tǒng)計分析。結(jié)果:空白對照組小鼠W0及W9時,CK1和CK10的表達(dá)差異均無統(tǒng)計學(xué)意義(P均0.05)。UV損傷組小鼠W0和W9時,CK1和CK10的表達(dá)差異均有統(tǒng)計學(xué)意義(P均0.05)。且UV損傷組小鼠CK1和CK10的表達(dá)呈明顯的定位改變。結(jié)論:多次UV照射能導(dǎo)致小鼠模型中CK1和CK10的表達(dá)明顯上調(diào)且定位異常。
