皮膚鱗狀細胞癌組織Syndecan-1和乙酰肝素酶表達及臨床意義

【摘要】:目的近期研究顯示,Syndecan-1和乙酰肝素酶(heparanase,HPA-1)可能參與多種惡性腫瘤的侵襲和轉(zhuǎn)移,但關(guān)于兩者與皮膚鱗狀細胞癌(cutaneous squamous cell carcinoma,CSCC)的關(guān)系較少報道。本研究旨在探討Syndecan-1和HPA-1在CSCC組織中的表達及其與臨床病理特征的關(guān)系。方法應(yīng)用免疫組化方法、蛋白質(zhì)印跡法和實時熒光定量PCR技術(shù)檢測2009-09-01-2014-03-31濰坊市人民醫(yī)院皮膚科93例CSCC及30名正常表皮中Syndecan-1和HPA-1蛋白及mRNA的表達水平。結(jié)果正常表皮、原位鱗癌、高分化鱗癌、中分化鱗癌和低分化鱗癌中Syndecan-1強陽性率分別為70.00%(21/30)、52.28%(11/21)、37.04%(10/27)、23.33%(7/30)和6.67%(1/15),CSCC Syndecan-1表達強度顯著低于正常表皮對照組,χ2=13.17,P0.01;在不同腫瘤厚度和分化程度的鱗癌中,SDC1表達強度隨腫瘤厚度的增加(χ2=11.66,P0.01)和分化程度的降低(χ2=12.51,P0.01)而有下降趨勢;CSCC患者中伴淋巴結(jié)轉(zhuǎn)移組Syndecan-1表達強度顯著低于無淋巴結(jié)轉(zhuǎn)移組,χ2=5.78,P0.05。CSCC組織Syndecan-1mRNA和蛋白表達水平顯著低于正常對照組,且隨著組織學分級的降低而依次降低(rs值分別為0.829和0.644,均P0.001)。正常表皮、原位鱗癌、高分化鱗癌、中分化鱗癌和低分化鱗癌中HPA-1陽性率分別為3.33%(1/30)、38.10%(8/21)、62.96%(15/27)、66.67%(20/30)和86.67%(13/15),CSCC中HPA-1表達強度顯著高于對照組,χ2=29.52,P0.01。在不同腫瘤厚度和分化程度的鱗癌中,HPA-1表達強度隨腫瘤厚度的增加(χ2=4.70,P0.05)和分化程度的降低(χ2=9.15,P0.05)而有增加的趨勢,CSCC患者中伴淋巴轉(zhuǎn)移組HPA-1表達強度(80.95%)顯著高于無淋巴結(jié)轉(zhuǎn)移組(54.17%),χ2=4.87,P0.05。CSCC組織HPA-1mRNA和蛋白表達水平顯著高于正常對照組,且隨著組織學分級的降低而依次升高(rs值分別為-0.793和-0.718,均P0.001)。CSCC中Syndecan-1與HPA-1表達呈負相關(guān),rs=-0.473,P0.01。結(jié)論 Syndecan-1表達降低與HPA-1表達增強可能促進CSCC的侵襲和轉(zhuǎn)移。
[Abstract]:Aim recent studies have shown that Syndecan-1 and heparanase (heparanase,HPA-1) may be involved in the invasion and metastasis of various malignant tumors, but there are few reports about their relationship with (cutaneous squamous cell carcinoma,CSCC in cutaneous squamous cell carcinoma (CSCC). The purpose of this study was to investigate the expression of Syndecan-1 and HPA-1 in CSCC and its relationship with clinicopathological features. Methods Immunohistochemistry was used. Protein blot and real-time fluorescence quantitative PCR were used to detect the expression of Syndecan-1, HPA-1 and mRNA in 93 cases of CSCC and 30 normal epidermis of Weifang people's Hospital. Results the strong positive rates of Syndecan-1 in normal epidermis, squamous cell carcinoma in situ, well differentiated squamous cell carcinoma, moderately differentiated squamous cell carcinoma and poorly differentiated squamous cell carcinoma were 70.00% (21 / 30), 52.28% (11 / 21), 37.04% (10 / 27), respectively. The expression intensity of 23. 33% (7? 30) and 6. 67% (1? 15), CSCC Syndecan-1) was significantly lower than that of the normal epidermis control group (蠂 2, 13. 17, P 0. 01). In squamous cell carcinoma with different tumor thickness and differentiation degree, the expression intensity of SDC1 decreased with the increase of tumor thickness (蠂 ~ 2 / 11.66, P0.01) and the degree of differentiation (蠂 ~ 2 / 12.51, P 0.01). The expression of Syndecan-1 in CSCC patients with lymph node metastasis was significantly lower than that in patients without lymph node metastasis, and the expression level of Syndecan-1mRNA and protein in CSCC tissues was significantly lower than that in normal controls (蠂 2 = 5.78, P0.05.CSCC, P < 0.05). And with the decrease of histologic grade (RS = 0.829 and 0.644, all P 0.001), the histological grade decreased with the decrease of the grade of histology (RS = 0.829 and 0.644, P 0.001). The positive rates of HPA-1 in normal epidermis, squamous cell carcinoma in situ, well differentiated squamous cell carcinoma, moderately differentiated squamous cell carcinoma and poorly differentiated squamous cell carcinoma were 3.33% (1 / 30), 38.10% (8 / 21), 62.96% (15 / 27), respectively. The expression intensity of HPA-1 in 66.67% (20 / 30) and 86.67% (13 / 15), CSCC) was significantly higher than that in the control group (蠂 2, 29.52, P 0.01). In squamous cell carcinoma with different tumor thickness and differentiation degree, the expression intensity of HPA-1 increased with the increase of tumor thickness (蠂 ~ 2 / 4.70, P 0.05) and the degree of differentiation (蠂 ~ 2 / 9.15, P 0.05), and the expression intensity increased with the increase of tumor thickness (蠂 ~ 2 / 4.70, P < 0.05). The expression of HPA-1 in CSCC patients with lymph node metastasis (80.95%) was significantly higher than that in patients without lymph node metastasis (54.17%). The expression of HPA-1mRNA and protein in P0.05.CSCC tissue was significantly higher than that in normal controls (蠂 2 = 4.87, P 0.05.CSCC). The expression of Syndecan-1 was negatively correlated with the expression of HPA-1, and the expression of rs=-0.473,P0.01. was negatively correlated with the decrease of histological grade (s =-0.793 and-0.718, P 0.001). Conclusion the decreased expression of Syndecan-1 and increased expression of HPA-1 may promote the invasion and metastasis of CSCC.
