p16INK4a和p14ARF在皮膚基底細胞癌和鱗狀細胞癌中的表達及其臨床意義

【摘要】：目的:基因突變在腫瘤的發(fā)病機制中起著重要作用,本文旨在探討pl6INK4a和p14ARF在皮膚基底細胞癌(BCC)和鱗狀細胞癌(SCC)中的表達特征及其臨床意義。 方法:采用免疫組織化學(xué)S-P法對25例皮膚基底細胞癌(BCC)、18例皮膚鱗狀細胞癌(SCC)標本和20例正常皮膚標本進行pl6INK4a和p14ARF蛋白的檢測,通過光學(xué)顯微鏡觀察免疫組織化學(xué)表達情況并應(yīng)用統(tǒng)計學(xué)方法對結(jié)果進行分析。 結(jié)果:pl6INK4a和p14ARF蛋白染色陽性產(chǎn)物都定位于細胞核內(nèi)。①pl6INK4a蛋白在BCC和SCC組的陽性表達率顯著低于正常皮膚組,差異有統(tǒng)計學(xué)意義(P0.05)。②p14ARF蛋白在BCC和SCC組的陽性表達率顯著低于正常皮膚組,差異有統(tǒng)計學(xué)意義( p0.05)。③pl6INK4a和p14ARF蛋白陽性表達率在BCC和SCC組及正常組織中無明顯相關(guān)性(p0.05)。 結(jié)論: pl6INK4a和p14ARF在皮膚BCC和SCC中的低表達,提示pl6INK4a和p14ARF基因突變在皮膚BCC和SCC的發(fā)生、發(fā)展中起著重要作用,有可能成為一種良好的皮膚腫瘤標志物。
[Abstract]:Aim: gene mutations play an important role in the pathogenesis of tumors. The aim of this study was to investigate the expression characteristics and clinical significance of pl6INK4a and p14ARF in (BCC) and (SCC) in cutaneous basal cell carcinoma (BCC) and squamous cell carcinoma (SCC). Methods: immunohistochemical method was used to detect pl6INK4a and p14ARF protein in 25 cases of basal cell carcinoma (BCC), 18 cases of squamous cell carcinoma of skin (SCC) and 20 cases of normal skin. The immunohistochemical expression was observed by optical microscope and the results were analyzed by statistical method. Results: the positive staining products of pl6INK4a and p14ARF protein were located in nucleus. 1the positive expression rate of pl6INK4a protein in BCC and SCC group was significantly lower than that in normal skin group. The positive expression rate of p14ARF protein in BCC and SCC group was significantly lower than that in normal skin group (P0.05). There was no significant correlation between the positive expression of pl6INK4a and p14ARF protein in BCC and SCC group and normal tissues (p0.05). Conclusion: the low expression of pl6INK4a and p14ARF in skin BCC and SCC suggests that pl6INK4a and p14ARF gene mutations play an important role in the occurrence and development of BCC and SCC and may be a good marker of skin tumor.
