p16INK4a和p14ARF在皮膚基底細(xì)胞癌和鱗狀細(xì)胞癌中的表達(dá)及其臨床意義

【摘要】:目的:基因突變?cè)谀[瘤的發(fā)病機(jī)制中起著重要作用,本文旨在探討pl6INK4a和p14ARF在皮膚基底細(xì)胞癌(BCC)和鱗狀細(xì)胞癌(SCC)中的表達(dá)特征及其臨床意義。 方法:采用免疫組織化學(xué)S-P法對(duì)25例皮膚基底細(xì)胞癌(BCC)、18例皮膚鱗狀細(xì)胞癌(SCC)標(biāo)本和20例正常皮膚標(biāo)本進(jìn)行pl6INK4a和p14ARF蛋白的檢測(cè),通過(guò)光學(xué)顯微鏡觀察免疫組織化學(xué)表達(dá)情況并應(yīng)用統(tǒng)計(jì)學(xué)方法對(duì)結(jié)果進(jìn)行分析。 結(jié)果:pl6INK4a和p14ARF蛋白染色陽(yáng)性產(chǎn)物都定位于細(xì)胞核內(nèi)。①pl6INK4a蛋白在BCC和SCC組的陽(yáng)性表達(dá)率顯著低于正常皮膚組,差異有統(tǒng)計(jì)學(xué)意義(P0.05)。②p14ARF蛋白在BCC和SCC組的陽(yáng)性表達(dá)率顯著低于正常皮膚組,差異有統(tǒng)計(jì)學(xué)意義( p0.05)。③pl6INK4a和p14ARF蛋白陽(yáng)性表達(dá)率在BCC和SCC組及正常組織中無(wú)明顯相關(guān)性(p0.05)。 結(jié)論: pl6INK4a和p14ARF在皮膚BCC和SCC中的低表達(dá),提示pl6INK4a和p14ARF基因突變?cè)谄つwBCC和SCC的發(fā)生、發(fā)展中起著重要作用,有可能成為一種良好的皮膚腫瘤標(biāo)志物。
[Abstract]:Aim: gene mutations play an important role in the pathogenesis of tumors. The aim of this study was to investigate the expression characteristics and clinical significance of pl6INK4a and p14ARF in (BCC) and (SCC) in cutaneous basal cell carcinoma (BCC) and squamous cell carcinoma (SCC). Methods: immunohistochemical method was used to detect pl6INK4a and p14ARF protein in 25 cases of basal cell carcinoma (BCC), 18 cases of squamous cell carcinoma of skin (SCC) and 20 cases of normal skin. The immunohistochemical expression was observed by optical microscope and the results were analyzed by statistical method. Results: the positive staining products of pl6INK4a and p14ARF protein were located in nucleus. 1the positive expression rate of pl6INK4a protein in BCC and SCC group was significantly lower than that in normal skin group. The positive expression rate of p14ARF protein in BCC and SCC group was significantly lower than that in normal skin group (P0.05). There was no significant correlation between the positive expression of pl6INK4a and p14ARF protein in BCC and SCC group and normal tissues (p0.05). Conclusion: the low expression of pl6INK4a and p14ARF in skin BCC and SCC suggests that pl6INK4a and p14ARF gene mutations play an important role in the occurrence and development of BCC and SCC and may be a good marker of skin tumor.
