寧夏地區(qū)回族和漢族白癜風(fēng)患者與HLA-DRB1的相關(guān)性探討

【摘要】:目的探究寧夏地區(qū)回漢族白癜風(fēng)患者與HLA-DRB1等位基因的相關(guān)性,以及回漢族人群在HLA-DRB1等位基因頻率分布的差異性,為揭示寧夏回漢族人群白癜風(fēng)發(fā)病機(jī)理及遺傳特點(diǎn)提供可靠實(shí)驗(yàn)室依據(jù)。 方法采用序列特異性引物-聚合酶鏈反應(yīng)(PCR-SSP)方法,對153例白癜風(fēng)患者及198例對照組進(jìn)行HLA-DRB1等位基因檢測,使用SPSSl3.0統(tǒng)計(jì)軟件,顯著性用χ2檢驗(yàn),P0.05認(rèn)為有顯著性差異。 結(jié)果①所檢測的HLA-DRB1的12個(gè)等位基因在寧夏地區(qū)回族正常人與漢族正常人的分布中無統(tǒng)計(jì)學(xué)差異。②寧夏漢族白癜風(fēng)患者較漢族正常對照組HLA-DRB1*09基因頻率顯著增高(9.2%vs2.5%),RR=4.222,χ2=9.787,Pc0.05。③寧夏漢族泛發(fā)型白癜風(fēng)患者較漢族正常對照組HLA-DRB1*09基因頻率顯著增高(9.4%vs2.5%),RR=4.308,χ2=9.390, Pc0.05。④寧夏回族白癜風(fēng)患者較回族正常對照組HLA-DRB1*16基因頻率顯著增高(10%vs1.3%),RR=9.750,χ2=9.255, Pc0.05。⑤寧夏回族泛發(fā)型白癜風(fēng)患者較漢族正常對照組HLA-DRB1*16基因頻率顯著增高(11.4%vs1.3%),RR=11.556,χ2=10.274, Pc0.05。⑥寧夏回漢族局限型白癜風(fēng)患者較回漢族正常對照組相比無統(tǒng)計(jì)學(xué)意義。 結(jié)論HLA-DRB1*09可能是寧夏漢族白癜風(fēng)患者尤其是泛發(fā)型患者的易感基因,或與易感基因相連鎖。HLA-DRB1*16可能是寧夏回族白癜風(fēng)患者尤其是回族泛發(fā)型患者的易感基因,或與易感基因相連鎖。
[Abstract]:Objective to investigate the association between HLA-DRB1 allele and vitiligo in Hui Han population in Ningxia and the difference of frequency distribution of HLA-DRB1 allele in Hui Han population. To provide a reliable laboratory basis for revealing the pathogenesis and genetic characteristics of vitiligo in Ningxia Hui Han population. Methods sequence specific primer polymerase chain reaction (PCR-SSP) was used to detect HLA-DRB1 alleles in 153 patients with vitiligo and 198 controls. The statistical software SPSSl3.0 was used and the significance was tested by 蠂 2 test. There was significant difference between the two groups (P0.05). Results 1 there was no significant difference in the distribution of 12 alleles of HLA-DRB1 between Hui and Han people in Ningxia area. 2 the frequency of HLA-DRB1*09 gene in Ningxia Han nationality patients with vitiligo was higher than that in Han nationality control group. The rate was significantly higher (9.2% vs 2.5%), The frequency of HLA-DRB1*09 gene in Ningxia Han nationality patients with generalized vitiligo was significantly higher than that in Han control group (9.4% vs 2.5%) and RR=4.308, 蠂 2 / 9.390,9.787, RR=4.222, 蠂 2 / 9.787, Pc0.05.3, P < 0.05, P < 0.01, respectively. The frequency of HLA-DRB1*16 gene in Ningxia Hui patients with vitiligo in Pc0.05.4 was significantly higher than that in the control group (10% vs 1.3%), and RR=9.750, 蠂 2 was 9.255, The frequency of HLA-DRB1*16 gene in Pc0.05.5 Hui nationality patients with generalized vitiligo in Ningxia was significantly higher than that in Han control group (11.4% vs 1.3%), and RR=11.556, 蠂 2 was 10.274, There was no significant difference in Pc0.05.6 between Ningxia Hui Han nationality patients with localized vitiligo and Hui Han normal controls. Conclusion Hla-DRB1*16 may be a susceptible gene in Ningxia Han nationality patients with vitiligo, especially in patients with generalized vitiligo. Hla-DRB1*16 may be a susceptible gene in Hui patients with vitiligo, especially those with generalized type of vitiligo in Ningxia. HLA-DRB1*09-DRB1*16 may be a susceptible gene in Hui patients with vitiligo, especially in patients with generalized vitiligo. Or linked to susceptible genes.
