不同條件下銀屑病患者表皮干細(xì)胞分離、培養(yǎng)、鑒定及生物學(xué)特性的初步研究

【摘要】:目的:不同條件下,對銀屑病患者表皮干細(xì)胞(Epidermal Stem Cells,ESCs)體外進(jìn)行分離、純化、培養(yǎng)及鑒定,,探索找出最佳培養(yǎng)體系,并對銀屑病患者表皮干細(xì)胞生物學(xué)特性進(jìn)行初步研究,為銀屑病皮損形成的深入研究提供依據(jù)。 方法:銀屑病皮損標(biāo)本37例、正常人皮膚標(biāo)本15例。ESCs分離、篩選、培養(yǎng):①Dispase酶、胰酶分別4℃過夜或37℃3小時四種消化條件隨機選擇,進(jìn)行真表皮分離效率比較;②制備單細(xì)胞懸液,比較單位面積銀屑病皮損與正常皮膚表皮細(xì)胞總數(shù)、活細(xì)胞數(shù);③IV型膠原不同濃度、不同黏附時間黏附法分選表皮干細(xì)胞,計算表皮細(xì)胞黏附率;④倒置顯微鏡下觀察細(xì)胞生長情況。⑤ESCs表面標(biāo)志物檢測:a.免疫熒光細(xì)胞化學(xué)法檢測原代12h、6d、12d的ESCs角蛋白19(K19)、角蛋白10(K10)的表達(dá);b.流式細(xì)胞儀檢測初黏附細(xì)胞、原代12天、P1代7天ESCs CD29(整合素β1)、CD71、P63的表達(dá)。細(xì)胞計數(shù)法繪制ESCs細(xì)胞生長曲線。 結(jié)果:ESCs分離、篩選、培養(yǎng):①2.5g/L Dispase II酶5%CO2、4℃過夜條件下真表皮分離效率高,總高效率達(dá)100%。②單位面積的銀屑病皮損表皮細(xì)胞的數(shù)量及活細(xì)胞量均大于正常皮膚標(biāo)本,結(jié)果具有顯著差異(P<0.05)。③a.IV型膠原黏附表皮細(xì)胞10min,可獲得純度較高的ESCs;b.50mg/L與100mg/L濃度IV型膠原表皮細(xì)胞黏附率未見顯著性差異(P>0.05)。④倒置顯微鏡下,銀屑病ESCs三到四天后開始快速生長,單個銀屑病ESCs形成的最大克隆遠(yuǎn)遠(yuǎn)大于正常皮膚組,培養(yǎng)數(shù)天后,銀屑病ESCs易于分化。⑤ESCs表面標(biāo)志物檢測:a.免疫熒光細(xì)胞化學(xué)法檢測:K19顯示為亮熒光綠色顆粒沉積在細(xì)胞表面及胞漿中,陽性表達(dá);K10未見熒光顆粒沉積;b.流式細(xì)胞儀檢測CD29、P63高表達(dá),CD71低表達(dá),結(jié)合免疫熒光細(xì)胞法,認(rèn)定所培養(yǎng)細(xì)胞為表皮干細(xì)胞。細(xì)胞計數(shù)法繪制ESCs生長曲線,結(jié)果顯示銀屑病ESCs短時間內(nèi)細(xì)胞倍增速度快。 結(jié)論:①成功分離、篩選、培養(yǎng)銀屑病ESCs,條件如下:2.5g/L Dispase II酶4℃過夜消化分離真表皮,2.5g/L胰蛋白酶消化制細(xì)胞懸液,50mg/L IV型膠原黏附10min篩選,可成功分選穩(wěn)定生長的ESCs;②銀屑病皮損表皮細(xì)胞數(shù)量、ESCs增殖能力、細(xì)胞生長形態(tài)、與正常皮膚存在差異;③銀屑病ESCs單細(xì)胞克隆能力強,生長迅速,易于分化。
