【摘要】：目的:觀察皮膚鱗狀細(xì)胞癌(SCC)和正常皮膚組織和細(xì)胞中抗凋亡蛋白cFLIP的表達(dá)情況,探討其與SCC臨床病理特征的關(guān)系及意義。 方法:采用免疫組織化學(xué)親和素-生物素-過氧化物酶(Avidin-Biotin-Peroxidase Complex,SABC)法檢測52例皮膚鱗狀細(xì)胞癌皮損和13例正常對照皮膚組織中cFLIP表達(dá);同時用逆轉(zhuǎn)錄PCR(RT-PCR)和免疫印跡(western-blot)方法檢測人皮膚鱗癌細(xì)胞系A(chǔ)431和永生化角質(zhì)形成細(xì)胞系Hacat中cFLIP各自的表達(dá)情況。 結(jié)果:皮膚鱗狀細(xì)胞癌組織中cFLIP表達(dá)的陽性率為67.31%,而正常皮膚組織中的陽性率為15.38%,兩者表達(dá)有顯著性差異(P0.01);cFLIP表達(dá)與病理分級有無淋巴結(jié)轉(zhuǎn)移相關(guān),與性別年齡病程發(fā)病部位無關(guān);cFLIP和細(xì)胞性增殖核抗原(PCNA)在皮膚鱗狀細(xì)胞癌中的表達(dá)具有相關(guān)性(r=0.781,P0.01);人皮膚鱗癌細(xì)胞系A(chǔ)431中cFLIP的mRNA和蛋白表達(dá)水平均高于永生化角質(zhì)形成細(xì)胞系Hacat。 結(jié)論:cFLIP在皮膚鱗狀細(xì)胞癌中表達(dá)顯著高于正常皮膚,cFLIP與腫瘤的病理學(xué)分級有無淋巴結(jié)轉(zhuǎn)移相關(guān),且和PCNA具有相關(guān)性;可能在皮膚鱗狀細(xì)胞癌的發(fā)生發(fā)展中發(fā)揮重要作用。。
[Abstract]:Objective: to investigate the expression of antiapoptotic protein (cFLIP) in (SCC) and normal skin tissues and cells of squamous cell carcinoma of skin (SCC), and to explore the relationship between the expression and clinicopathological features of SCC. Methods: immunohistochemical avidin-biotin-peroxidase (Avidin-Biotin-Peroxidase Complex,SABC) method was used to detect the expression of cFLIP in 52 cases of cutaneous squamous cell carcinoma and 13 cases of normal control. The expression of cFLIP in human cutaneous squamous cell carcinoma cell line A431 and immortalized keratinocyte cell line Hacat was detected by reverse transcription PCR (RT-PCR) and Western blot (western-blot). Results: the positive rate of cFLIP expression was 67.31 in squamous cell carcinoma of skin and 15.38 in normal skin tissue. There was significant difference between them (P0.01); cFLIP expression and pathological grade). Is there a lymph node metastasis associated with sex? Age? Course? The expression of cFLIP and cellular proliferating nuclear antigen (PCNA) in squamous cell carcinoma of skin was correlated (r = 0.781P 0.01). The expression of cFLIP mRNA and protein in human cutaneous squamous cell carcinoma cell line A431 was higher than that in immortalized keratinocyte cell line Hacat.. Conclusion: the expression of cFLIP in cutaneous squamous cell carcinoma is significantly higher than that in normal skin. Lymph node metastasis is associated with PCNA, which may play an important role in the development of cutaneous squamous cell carcinoma.
