磷脂酰肌醇3激酶和蛋白激酶B在小兒血管瘤中的表達(dá)及意義

【摘要】:目的:探討磷脂酰肌醇3激酶(P13K)、蛋白激酶B(Akt)在小兒血管瘤的發(fā)生、發(fā)展及消退過程中的作用。 方法:收集遵義醫(yī)學(xué)院附屬醫(yī)院2009.01-2012.03住院,未經(jīng)其他治療、單純手術(shù)切除并經(jīng)病理診斷為血管瘤的標(biāo)本,合Mulliken分類法與增殖細(xì)胞核抗原(PCNA)的表達(dá)對(duì)血管瘤進(jìn)行分類和分期,抽取增生期與消退期血管瘤各20例,采用免疫組化檢測血管瘤組織中磷脂酰肌醇3激酶和蛋白激酶B的表達(dá)水平,并進(jìn)行圖像分析。采用定量PCR檢測血管瘤組織中磷脂酰肌醇3激酶和蛋白激酶B的(?)nRNA表達(dá)情況。 結(jié)果: 1.20例增殖期血管瘤均表達(dá)PI3Kp85、p-Akt,20例退化期血管瘤中PI3Kp85、 p-Akt的表達(dá)率分別為14例(70%)、12例(60%),差異均有統(tǒng)計(jì)學(xué)意義(P0.05);增殖期血管瘤PI3Kp85(?)勺表達(dá)強(qiáng)度為1例(+)、7例(++)、12例(+++),退化期血管瘤PI3Kp85(?)勺表達(dá)強(qiáng)度為6例(-)、8例(+)、5例(++)、1例(+++),統(tǒng)計(jì)學(xué)差異較大(P0.01);增殖期血管瘤p-Akt的表達(dá)強(qiáng)度為2例(+)、11例(++)、7例(+++),退化期血管瘤p-Akt的表達(dá)強(qiáng)度為8例(-)、9例(+)、3例(++),統(tǒng)計(jì)學(xué)差異較大(P0.01)。 2.20例增殖期血管瘤PI3Kp85、p-Akt的積分光密度分別為191.23±40.45、178.73±38.49,退化期血管瘤PI3Kp85、P-Akt的積分光密度分別為46.63±13.08、64.93±21.22,統(tǒng)計(jì)學(xué)差異均較大(P0.01)。PI3Kp85與p-Akt的表達(dá)之間呈正相關(guān)關(guān)系(r=0.553,P0.05)。 3.20例增殖期血管瘤PI3Kp85mRNA、p-AktmRNA的相對(duì)表達(dá)值分別為0.0807±0.0136、0.0841±0.0137,退化期血管瘤PI3Kp85、P-Akt的積分光密度分別為0.0203±0.0098、0.0324±0.0178,統(tǒng)計(jì)學(xué)差異均較大(P0.01)。PI3Kp85mRNA、 P-AktmRNA的表達(dá)之間呈正相關(guān)關(guān)系(r=0.5503P0.01)。 結(jié)論:小兒增生期和消退期血管瘤組織中PI3Kp85、p-Akt及其mRNA表達(dá)有差異;PI3K/Akt信號(hào)通路可能在小兒血管瘤的病理演變中發(fā)揮作用。
[Abstract]:Objective: to investigate the role of phosphatidylinositol 3 kinase (P13 K) and protein kinase (B (Akt) in the occurrence, development and regression of children hemangioma. Methods: the specimens of hemangioma were collected from the affiliated Hospital of Zunyi Medical College 2009.01-2012.03. Mulliken classification and proliferating cell nuclear antigen (PCNA) expression were used to classify and stage hemangiomas. The expression levels of phosphatidylinositol 3 kinase and protein kinase B in hemangioma tissues were detected by immunohistochemistry and image analysis. The expression of phosphatidylinositol 3 kinase and protein kinase B nRNA was detected by quantitative PCR. Results: 1.The expression rates of PI3Kp85, p-Akt in proliferative hemangioma were 14 cases (70%) and 12 cases (60%), respectively. The difference was statistically significant (P0.05). Proliferative hemangioma PI3Kp85 (?) 1 case of (), 7 cases of (), 12 cases of (), degeneration hemangioma PI3Kp85 (?) The expression intensity of dipper was 6 cases (-), 8 cases of (), was 5 cases of (), 1 case of (), was significantly different (P0.01). The expression intensity of p-Akt in proliferative hemangioma was 2 cases of (), 11 cases of (), 7 cases of (), degenerative hemangioma 8 cases (-) and 9 cases of (), 3 cases of (),. The statistical difference was significant (P0.01). 2. The integral optical density of PI3Kp85,p-Akt was 191.23 鹵40.45178.73 鹵38.49 in proliferative hemangioma and 46.63 鹵13.08 鹵64.93 鹵21.22 in degenerative hemangioma, respectively. There were significant statistical differences (P0.01). There was a positive correlation between the expression of PI3Kp85 and p-Akt (P 0.05). 3. The relative expression of PI3Kp85mRNA,p-AktmRNA in proliferative hemangioma was 0.0807 鹵0.0136 鹵0.0137, and the integral optical density of PI3Kp85,P-Akt in degenerative hemangioma was 0.0203 鹵0.0098 鹵0.0324 鹵0.0178, respectively. There were significant statistical differences (P0.01). There was a positive correlation between the expression of PI3Kp85mRNA, P-AktmRNA (r=0.5503P0.01). Conclusion: there are differences in expression of PI3Kp85,p-Akt and mRNA between proliferative and regressive hemangiomas in children, and PI3K/Akt signaling pathway may play a role in the pathological evolution of hemangioma in children.
