Cyr61上調(diào)人角質(zhì)形成細(xì)胞系表達(dá)趨化因子與銀屑病發(fā)生機(jī)制初探

【摘要】:銀屑病是一種常見的慢性炎癥性皮膚疾病,多種炎癥細(xì)胞與炎癥因子參與其發(fā)病。我們已有研究發(fā)現(xiàn)促炎因子Cyr61/CCN1通過活化角質(zhì)細(xì)胞而加劇皮損,但是否還有其他促炎機(jī)制尚不清楚。本研究通過Cyr61介導(dǎo)角質(zhì)細(xì)胞產(chǎn)生趨化因子的體外實(shí)驗(yàn),探討Cyr61參與銀屑病發(fā)生新途徑。采用Real-time PCR方法檢測(cè)人角質(zhì)形成細(xì)胞系(HaCat細(xì)胞)在外源性Cyr61作用下趨化因子的表達(dá)格局。實(shí)驗(yàn)結(jié)果顯示Cyr61能夠在體外促進(jìn)人角質(zhì)形成細(xì)胞系表達(dá)多種趨化因子(如CCL2,CCL17,CCL22,CXCL1,CXCL10,CXCL11(P0.05)),提示Cyr61可能通過促進(jìn)角質(zhì)形成細(xì)胞系表達(dá)多種趨化因子從而參與DC,Th17,Th1等細(xì)胞的趨化。提示Cyr61可能通過促進(jìn)角質(zhì)形成細(xì)胞系表達(dá)趨化因子介導(dǎo)炎性細(xì)胞局部浸潤(rùn)增多,加劇銀屑病發(fā)病。
[Abstract]:Psoriasis is a common chronic inflammatory skin disease. Many inflammatory cells and inflammatory factors are involved in the pathogenesis of psoriasis. We have found that Cyr61/CCN1 exacerbates skin lesions by activating keratinocytes, but it is not clear whether there are other inflammatory mechanisms. In this study, Cyr61 mediated keratinocytes to produce chemokines in vitro to explore the role of Cyr61 in the pathogenesis of psoriasis. The expression pattern of chemokines in human keratinocytes (HaCat cells) induced by exogenous Cyr61 was detected by Real-time PCR method. The results show that Cyr61 can promote the expression of many chemokines (such as CCL2,CCL17,CCL22,CXCL1,CXCL10,CXCL11 (P0.05),) in human keratinocytes in vitro. These results suggest that Cyr61 may be involved in the chemotaxis of DC,Th17,Th1 and other cells by promoting the expression of many chemokines in keratinocytes. It is suggested that Cyr61 may increase the local infiltration of inflammatory cells by promoting the expression of chemokines in keratinocytes and aggravate the pathogenesis of psoriasis.
