【摘要】：目的:研究Wnt10b對(duì)體外培養(yǎng)小鼠觸須毛囊生長的作用。方法:構(gòu)建Dct-Lac Z小鼠觸須毛囊體外培養(yǎng)模型。用腺病毒Ad-Wnt10b和Ad-GFP感染觸須毛囊2 d、4 d并對(duì)觸須生長長度進(jìn)行測量。X-gal染色檢測觸須毛囊中黑素細(xì)胞數(shù)量。Western blot檢測酪氨酸相關(guān)蛋白1(tyrosinase-related protein 1,TRP-1)、酪氨酸酶(tyrosinase,TYR)蛋白的表達(dá)水平。結(jié)果:腺病毒感染4 d后長度測量結(jié)果顯示W(wǎng)nt10b處理組的觸須毛囊生長長度明顯高于GFP對(duì)照組(0.58±0.03 vs.0.09±0.05,P=0.000);X-gal染色結(jié)果顯示W(wǎng)nt10b處理組的黑素細(xì)胞數(shù)量明顯多于GFP對(duì)照組;Western blot結(jié)果顯示W(wǎng)nt10b處理組的觸須毛囊中TRP-1和TYR的蛋白表達(dá)明顯升高(1.14±0.16 vs.0.55±0.22,1.34±0.33 vs.0.77±0.09;P=0.020,P=0.045)。結(jié)論:Wnt10b能夠促進(jìn)小鼠觸須毛囊的體外生長。
[Abstract]:Objective: to study the effect of Wnt10b on the growth of mouse hair follicles in vitro. Methods: Dct-Lac Z mouse hair follicles were cultured in vitro. The hair follicles were infected with adenovirus Ad-Wnt10b and Ad-GFP for 2 d and the length of the hair follicles was measured. The number of melanocytes in the hair follicles was detected by X-gal staining. The expression of tyrosine associated protein 1 (tyrosinase-related protein 1) and tyrosinase (tyrosinase,TYR) protein in the hair follicles was detected by. Western blot. Results: the length measurement after 4 days of adenovirus infection showed that the growth length of hair follicles in Wnt10b treated group was significantly longer than that in GFP control group (0.58 鹵0.03 鹵0.05 vs.0.09 鹵0.05 P0. 000), and X-gal staining showed that the number of melanocytes in Wnt10b treated group was significantly larger than that in GFP control group. The expression of TRP-1 and TYR in the hair follicles of the Wnt10b treated group was significantly increased (1.14 鹵0.16 vs.0.55 鹵0.22 vs.0.55 鹵0.33 vs.0.77 鹵0.33 vs.0.77 鹵0.09 vs.0.77 鹵0.020 TYR 0.045). Conclusion: Wnt10b can promote the growth of mouse hair follicles in vitro.
