木天蓼對VEGF121刺激血管內(nèi)皮細(xì)胞分泌NO的影響

【摘要】:目的探討木天蓼對血管內(nèi)皮生長因子121(VEGF121)刺激血管內(nèi)皮細(xì)胞(HUVEC)產(chǎn)生一氧化氮(NO)的影響。方法體外培養(yǎng)HUVEC,分別用VEGF121,VEGF121+木天蓼及實(shí)驗(yàn)培養(yǎng)基進(jìn)行刺激,MTT檢測各實(shí)驗(yàn)組HUVEC的增殖活力,采用ELISA方法檢測培養(yǎng)基中NO的含量。結(jié)果 VEGF121能明顯刺激HUVEC分泌NO,與對照組相比差異有統(tǒng)計(jì)學(xué)意義(P=0.044);培養(yǎng)8h后,MTT檢測木天蓼+VEGF121組、VEGF121組與對照組相比HUVEC增殖活力差異無統(tǒng)計(jì)學(xué)意義(P0.05);木天蓼醇提取液(100mg/m L)可抑制VEGF121刺激HUVEC產(chǎn)生NO,差異有統(tǒng)計(jì)學(xué)意義(P=0.008)。結(jié)論VEGF121可促進(jìn)HUVEC分泌NO,而木天蓼對其具有抑制作用,該結(jié)果為木天蓼治療VEGF相關(guān)的皮膚疾病提供了基礎(chǔ)理論。
[Abstract]:Objective to investigate the effect of Polygonum hydropiper on the production of nitric oxide (NO) (NO) by vascular endothelial growth factor 121 (VEGF121)-stimulated (HUVEC) in vascular endothelial cells (VEC). Methods HUVECs were cultured in vitro. The proliferative activity of HUVEC in each experimental group was detected with VEGF121VEGF121 and experimental medium, and the content of no in culture medium was detected by ELISA method. Results VEGF121 significantly stimulated HUVEC to secrete no (P0.044), and after 8 hours culture, there was no significant difference in the proliferation activity of HUVEC between VEGF121 group and control group (P0.05). (100mg/m L) could inhibit the production of no in HUVEC stimulated by VEGF121 (P0. 008). Conclusion VEGF121 can promote the secretion of no by HUVEC, but Polygonum hydropiper can inhibit it. The results provide a basic theory for the treatment of VEGF related skin diseases by Polygonum hydropiper.
