【摘要】：目的:探究白細(xì)胞介素22(interleukin-22,IL-22)在細(xì)胞增殖以及細(xì)胞因子表達(dá)方面對人臍靜脈內(nèi)皮細(xì)胞(human umbilical vein endothelial cells,HUVECs)的影響。方法:用RT-PCR技術(shù)檢測IL-22R1在HUVECs中的表達(dá)情況。將以脂多糖(Lipopolysaccharide,LPS)處理后的HUVECs作為血管炎細(xì)胞模型,CCK8實(shí)驗(yàn)用于檢測IL-22作用前后HUVECs的增殖能力變化情況,q RT-PCR技術(shù)以及Western blot技術(shù)則分別用于檢測IL-22作用前后IL-6,IL-8,CC類趨化因子配體2(chemokine motif lig and 2,CCL2),細(xì)胞間粘附分子1(intercellular cell adhesion molecule-1,ICAM1),血管細(xì)胞粘附分子-1(vascular cell adhesion molecule 1,VCAM1),趨化因子受體4(chemokine receptor 4,CXCR4)和CCL20等細(xì)胞因子在HUVECs中mRNA水平以及蛋白質(zhì)水平的表達(dá)變化情況。結(jié)果:實(shí)驗(yàn)結(jié)果顯示IL-22R1 mRNA在HUVECs中廣泛表達(dá)。IL-22作用于HUVECs后,細(xì)胞的增殖率得到明顯提高。此外,LPS(脂多糖)可以顯著抑制HUVECs的增殖率,而被IL-22預(yù)處理過的HUVECs其增殖率不受LPS的抑制。在IL-22作用于HUVECs后,趨化因子CCL2和CCL20的表達(dá)在mRNA水平及蛋白質(zhì)水平均可見明顯增加,而炎癥因子IL-6及IL-8的表達(dá)則未見明顯變化。結(jié)論:IL-22的受體IL-22R1在HUVECS中廣泛表達(dá)。IL-22不僅可以提高HUVECs的增殖能力,還可以抑制由LPS誘導(dǎo)的增殖抑制。此外,IL-22還可以提高趨化因子CCL2與CCL20的表達(dá),卻對炎癥因子IL-6及IL-8的表達(dá)無明顯影響。由此可以推測IL-22在血管炎的發(fā)展過程中具有抗炎的作用。
[Abstract]:Aim: to investigate the effects of interleukin-22 (IL-22) on the proliferation and cytokine expression of human umbilical vein endothelial cells (human umbilical vein endothelial cells). Methods: RT-PCR technique was used to detect the expression of IL-22R1 in HUVECs. Using lipopolysaccharide (LPS) -treated HUVECs as a vasculitis cell model, CCK8 assay was used to detect the proliferation ability of HUVECs before and after IL-22 treatment. Q RT-PCR technique and Western blot technique were used to detect IL-6 and IL-8 CC chemokines before and after IL-22 treatment, respectively. The expression of cytokines such as ligand 2 (chemokine motif lig and 2 (CCL2), intercellular adhesion molecule 1 (intercellular cell adhesion molecule-1 (ICAM1), vascular cell adhesion molecule 1 (VCAM1), chemokine receptor 4 (CXCR4) and CCL20 in HUVECs were detected. Results: the results showed that the proliferation rate of HUVECs cells was significantly increased after the expression of. IL-22 was widely expressed in HUVECs. In addition, lipopolysaccharide (LPS) could significantly inhibit the proliferation of HUVECs, but HUVECs pretreated with IL-22 could not inhibit the proliferation of HUVECs by LPS. The expression of chemokine CCL2 and CCL20 increased significantly at the level of mRNA and protein, but the expression of IL-6 and IL-8, the inflammatory factors, did not change significantly after the action of IL-22 on HUVECs. Conclusion the expression of IL-22R1 in HUVECS can not only increase the proliferation ability of HUVECs, but also inhibit the proliferation inhibition induced by LPS. In addition, IL-22 could increase the expression of chemokine CCL2 and CCL20, but had no effect on the expression of IL-6 and IL-8. Therefore, it can be inferred that IL-22 plays an anti-inflammatory role in the development of vasculitis.
