HIF-1α和COX-2在銀屑病中的表達(dá)及相關(guān)性研究

【摘要】:第一部分HIF-lα與COX-2在銀屑病皮損中的表達(dá)及相關(guān)性研究 目的:檢測(cè)HIF-1α和COX-2在尋常型銀屑病皮損及正常人皮膚組織中的表達(dá)水平,探討HIF-1α和COX-2在銀屑病發(fā)病機(jī)理中的作用。 方法:詳細(xì)收集30例尋常型銀屑病患者和20例正常人的臨床資料,并收集皮損和皮膚組織;(1)用半定量RT-PCR檢測(cè)銀屑病皮損和正常皮膚組織中HIF-1α和COX-2 mRNA表達(dá)情況,(2)用Western Blot檢測(cè)銀屑病皮損和正常皮膚組織中HIF-1α和COX-2蛋白表達(dá)情況。 結(jié)果:HIF-1α和COX-2 mRNA相對(duì)表達(dá)量在尋常型銀屑病組(1.32±0.04和1.21±0.04)的表達(dá)顯著高于正常對(duì)照組(0.49±0.03和0.31±0.03),兩組相比差異有統(tǒng)計(jì)學(xué)意義(P0.05);且HIF-1αmRNA相對(duì)表達(dá)量與COX-2 mRNA相對(duì)表達(dá)量呈正相關(guān)(r=0.664,P0.05);HIF-1α和COX-2蛋白在尋常型銀屑病組(0.76±0.03和0.62±0.03)的表達(dá)顯著高于正常對(duì)照組(0.44±0.05和0.28±0.06),,兩組相比差異均有統(tǒng)計(jì)學(xué)意義(P0.05);且HIF-1α蛋白表達(dá)量與COX-2蛋白表達(dá)量呈正相關(guān)(r=0.601,P0.05)。 結(jié)論: HIF-1α和COX-2在銀屑病皮損中異常高表達(dá)且呈正相關(guān),可能在銀屑病的發(fā)生發(fā)展過程中起著重要作用。 第二部分HIF-1α和COX-2在豚鼠銀屑病樣模型皮損中的表達(dá)及相關(guān) 性研究 目的:研究HIF-1α和COX-2在豚鼠銀屑病樣動(dòng)物模型皮損表皮中的表達(dá)及相關(guān)性,探討其在銀屑病發(fā)病機(jī)制的作用。 方法:用5%普萘洛爾乳劑外涂豚鼠耳背部皮膚誘導(dǎo)銀屑病樣動(dòng)物模型,應(yīng)用免疫組化法檢測(cè)正常豚鼠表皮和豚鼠銀屑病樣動(dòng)物模型皮損表皮中HIF-1α和COX-2表達(dá)。 結(jié)果:5%普萘洛爾乳劑外涂所致豚鼠銀屑病樣動(dòng)物模型皮損中HIF-1α和COX-2表達(dá)水平較正常對(duì)照組明顯增高,差異有統(tǒng)計(jì)學(xué)意義(P0.01)。 結(jié)論:HIF-1α和COX-2在豚鼠銀屑病樣動(dòng)物模型皮損的表皮中表達(dá)明顯增高且呈正相關(guān);不僅在組織病理學(xué)而且在分子水平上與人類銀屑病皮損基本一致。
[Abstract]:Part I the expression of HIF-l 偽 and COX-2 in psoriatic lesions and their correlation objective: to detect the expression of HIF-1 偽 and COX-2 in skin tissues of psoriasis vulgaris and normal people, and to explore the role of HIF-1 偽 and COX-2 in the pathogenesis of psoriasis. Methods: the clinical data of 30 patients with psoriasis vulgaris and 20 normal subjects were collected in detail. (1) the expressions of HIF-1 偽 and COX-2 mRNA in psoriatic lesions and normal skin tissues were detected by semi-quantitative RT-PCR, and (2) the expression of HIF-1 偽 and COX-2 protein in psoriatic lesions and normal skin tissues were detected by Western Blot. Results the relative expression of 1: HIF-1 偽 and COX-2 mRNA in psoriasis vulgaris group (1.32 鹵0.04 and 1.21 鹵0.04) was significantly higher than that in normal control group (0.49 鹵0.03 and 0.31 鹵0.03), and the relative expression of HIF-1 偽 mRNA was positively correlated with the relative expression of COX-2 mRNA (P 0.05). The expression of HIF-1 偽 and COX-2 protein in psoriasis vulgaris group (0.76 鹵0.03 and 0.62 鹵0.03) was significantly higher than that in normal control group (0.44 鹵0.05 and 0.28 鹵0.06). Conclusion: the overexpression of HIF-1 偽 and COX-2 in psoriatic lesions is positively correlated and may play an important role in the development of psoriasis. The expression of HIF-1 偽 and COX-2 in the lesions of guinea-pig psoriasis model objective: to study the expression and correlation of HIF-1 偽 and COX-2 in the epidermis of psoriasis model of guinea pig. To explore its role in the pathogenesis of psoriasis. Methods: 5% propranolol emulsion was applied to induce psoriasis animal model. The expression of HIF-1 偽 and COX-2 in normal guinea pig epidermis and lesion epidermis of guinea-pig psoriasis model were detected by immunohistochemistry. Results the expression of HIF-1 偽 and COX-2 in the lesions of guinea pig psoriatic model induced by 5% propranolol emulsion was significantly higher than that in normal control group (P0.01). Conclusion the expressions of 1 / HIF-1 偽 and COX-2 in the epidermis of psoriasis like animal model of guinea pig were significantly increased and positively correlated with those of human psoriatic lesions, not only in histopathology but also at the molecular level.
