扁平苔蘚皮損中VEGF、p-Akt、Survivin表達(dá)與凋亡的關(guān)系

【摘要】: 目的:研究VEGF、p-Akt和Survivin蛋白在扁平苔蘚皮損中的表達(dá)情況以及與扁平苔蘚皮損中細(xì)胞凋亡的關(guān)系,探討PI3K/Akt信號(hào)通路和細(xì)胞凋亡在扁平苔蘚發(fā)病中的作用。 方法:采用免疫組織化學(xué)法檢測(cè)30例扁平苔蘚皮損及30例正常皮膚組織標(biāo)本中VEGF、p-Akt和Survivin的表達(dá)情況和表達(dá)部位。采用原位末端標(biāo)記技術(shù)(TUNEL)法檢測(cè)皮損處細(xì)胞凋亡的情況。并對(duì)VEGF、p-Akt和Survivin在皮損中的表達(dá)水平進(jìn)行相關(guān)性分析。 結(jié)果:VEGF、p-Akt和Survivin的表達(dá)均以細(xì)胞漿著黃色或棕黃色顆粒為陽(yáng)性,采用積分光密度(integrated optical density, IOD)值的大小表示陽(yáng)性表達(dá)強(qiáng)度。扁平苔蘚皮損中VEGF、p-Akt和Survivin的IOD值分別為13.584±1.131、14.405±1.263和10.537±1.212均明顯高于正常對(duì)照組(P0.01),并且三項(xiàng)指標(biāo)的表達(dá)水平呈正相關(guān)(P0.01)。扁平苔蘚組皮損處表皮角質(zhì)形成細(xì)胞的凋亡指數(shù)(apoptosis index, AI)為72.817±9.234,明顯高于正常對(duì)照組28.160±3.464(P0.01)。 結(jié)論:扁平苔蘚皮損表皮角質(zhì)形成細(xì)胞高表達(dá)的VEGF可能通過(guò)PI3K/Akt信號(hào)通路使Survivin的表達(dá)增高。PI3K/Akt信號(hào)通路可能參與了扁平苔蘚的發(fā)病。扁平苔蘚皮損處同時(shí)存在角質(zhì)形成細(xì)胞的過(guò)度增殖和凋亡亢進(jìn)。
[Abstract]:Aim: to investigate the expression of VEGFP-Akt and survivin protein in the lesions of lichen planus and the relationship between the expression of VEGFP-Akt and apoptosis in lichen planus and to explore the role of PI3K / Akt signaling pathway and apoptosis in the pathogenesis of lichen planus. Methods: the expression and location of VEGFP-Akt and survivin in 30 cases of lichen planus lesions and 30 cases of normal skin tissue were detected by immunohistochemical method. In situ end labeling (Tunel) was used to detect apoptosis in lesions. The expression levels of VEGF-Akt and survivin in skin lesions were analyzed. Results the positive expression of the cell cytoplasm was yellow or brown granules, and the intensity of the positive expression was indicated by the value of integrated optical density (integrated optical density,). The IOD values of VEGFP-Akt and survivin in lichen planus lesions were 13.584 鹵1.131, 14.405 鹵1.263 and 10.537 鹵1.212, respectively, which were significantly higher than those in the normal control group (P0.01), and the expression levels of the three indexes were positively correlated (P0.01). The apoptotic index (apoptosis index, AI) of epidermal keratinocytes in the lichen planus group was 72.817 鹵9.234, which was significantly higher than that in the normal control group (28.160 鹵3.464). Conclusion: the overexpression of VEGF in epidermal keratinocytes of lichen planus may increase the expression of survivin through PI3K / Akt signaling pathway. PI3K / Akt signaling pathway may be involved in the pathogenesis of lichen planus. Excessive proliferation and apoptosis of keratinocytes exist in the lesions of lichen planus.
