Toll樣受體2、4基因多態(tài)性與麻風(fēng)的相關(guān)性研究

【摘要】: 目的:研究Toll樣受體2(Toll-like receptor2, TLR2) (597T/C)和TLR4(896A/G)基因單核苷酸多態(tài)性(SNP)與麻風(fēng)的相關(guān)性,探討麻風(fēng)的遺傳易感性。方法:選擇中國貴州畢節(jié)地區(qū)麻風(fēng)病例117例[按類型分為瘤型(79例)和結(jié)核樣型(38例)、按集聚性分為家族集聚性(43例)和非家族集聚性(74例)],健康正常對(duì)照50例;用聚合酶鏈反應(yīng)限制性片斷長度多態(tài)性(PCR-RFLP)法分析各樣本TLR2 597T/C和TLR4 896A/G位點(diǎn)的基因型,比較麻風(fēng)和正常組基因型分布所占比率以及麻風(fēng)病例中各組基因型分別占有比率。SPSS16.0軟件的x2檢驗(yàn)對(duì)各組例數(shù)進(jìn)行統(tǒng)計(jì)處理。結(jié)果:TLR2(597T/C)TT、TC、CC三種基因型在麻風(fēng)患者和健康人中的分布頻率分別為46.2%、40.2%、13.6%和48.0%、42.0%、10.0%,無明顯差異(x2=0.431,P0.05);瘤型和結(jié)核樣TT、TC、CC基因型所占比率分別為44.3%、40.5%、15.2%和50.0%、39.5%、10.5%,差異無統(tǒng)計(jì)學(xué)意義(x2=0.595,P0.05);家族集聚性和非家族性TT、TC、CC基因型所占比率分別為48.8%、37.2%、14.0%和44.6%、41.9%、13.5%,差異無統(tǒng)計(jì)學(xué)意義(x2=0.258,P0.05))。所有研究對(duì)象未檢測(cè)到TLR4(896A/G)多態(tài)性,基因型均為896AA野生純合子。結(jié)論:TLR2(597T/C)、TLR4(896T/G)多態(tài)性與中國貴州畢節(jié)地區(qū)麻風(fēng)無相關(guān),TLR4(896A/G)多態(tài)性在中國人中罕見。
[Abstract]:Objective: to study the association between Toll-like receptor 2 (TLR2) (597T / C) and TLR4 (896A / G) gene single nucleotide polymorphism (SNP) in leprosy and to explore the genetic susceptibility of leprosy. Methods: 117 cases of leprosy in Bijie District, Guizhou Province, China were divided into tumor type (79 cases) and tuberculoid type (38 cases), familial agglomeration (43 cases) and non-familial agglomeration (74 cases), and healthy control group (50 cases). The genotypes of TLR2 597T / C and TLR4896A / G loci were analyzed by polymerase chain reaction restriction fragment length polymorphism (PCR-RFLP). The percentage of genotype distribution in leprosy group and normal group and the percentage of genotype in each group were compared. SPSS 16.0 software x2 test was used to analyze the number of cases in each group. Results the distribution frequency of TLR2 (597T / C) TCCC genotypes in leprosy patients and healthy persons was 46.2% and 48.2%, respectively, and there was no significant difference between them (x20.431P0.05), and the percentage of TCCCC genotypes in tumor-type and tuberculoid TTTCnC was 44.3%, 40.515.2% and 59.55.50%, respectively, and there was no significant difference between them (x20.595P0.05). The percentage of TCC-CC genotypes was 48.8% and 44.69%, respectively. There was no significant difference between them (x20.258%, P0.05). No TLR4 (896A / G) polymorphism was detected in all subjects and the genotype was 896AA wild homozygote. ConclusionTwo TLR2 (597T / C) TLR4 (896T / G) polymorphism is not associated with leprosy in Bijie region, Guizhou Province, China. The polymorphism of TLR4 (896A / G) is rare in Chinese.
