【摘要】：目的研究神經(jīng)鞘氨醇磷酸膽堿(shingosylphosphorylcholine,SPC)對體外培養(yǎng)人真皮成纖維細胞分泌基質(zhì)金屬蛋白酶-1(MMP-1)、基質(zhì)金屬蛋白酶-3(MMP-3)、基質(zhì)金屬蛋白酶內(nèi)源性組織抑制劑-1(TIMP-1)的影響,探索在促進皮膚創(chuàng)傷愈合方面SPC的作用機制。方法通過MTT的方法檢測SPC在成纖維細胞增殖效率方面的作用。利用RT-PCR和Western blotting的方法檢測體外培養(yǎng)人真皮成纖維細胞經(jīng)5μmol/L SPC誘導(dǎo)后,表達損傷修復(fù)相關(guān)蛋白MMP-1,MMP-3,TIMP-1的情況。結(jié)果一定濃度的SPC具有促進成纖維細胞增殖的作用。5μmol/L SPC具有上調(diào)成纖維細胞表達MMP-1,MMP-3,TIMP-1 m RNA和蛋白的作用(P0.05)。結(jié)論實驗證實體外培養(yǎng)人真皮成纖維細胞在經(jīng)SPC干預(yù)后,其表達細胞外基質(zhì)調(diào)節(jié)蛋白MMP-1,MMP-3,TIMP-1的作用得到增強,提示SPC具有促進皮膚創(chuàng)傷愈合的作用。
[Abstract]:Objective to study the effect of SPCs on the secretion of matrix metalloproteinase-1 (MMP-1), matrix metalloproteinase-3 (MMP)-3 and matrix metalloproteinase (MMP)-1 (TIMP-1) in human dermal fibroblasts in vitro, and to investigate the effects of SPCs on the secretion of matrix metalloproteinase-1 (MMP-1), matrix metalloproteinase-3 (MMP-2) and matrix metalloproteinase inhibitor (MMP-1) in vitro. To explore the mechanism of SPC in promoting skin wound healing. Methods the effect of SPC on the proliferation of fibroblasts was detected by MTT. RT-PCR and Western blotting were used to detect the expression of MMP-1, MMP-3 and TIMP-1 in cultured human dermal fibroblasts induced by 5 渭 mol/L SPC. Results A certain concentration of SPC could promote the proliferation of fibroblasts. 5 渭 mol/L SPC could up-regulate the expression of MMP-1, MMP-3, TIMP-1 RNA and protein in fibroblasts. Conclusion the results suggest that the expression of MMP-1, MMP-3 and TIMP-1 in cultured human dermal fibroblasts is enhanced after the intervention of SPC, suggesting that SPC can promote the healing of skin trauma.
