【摘要】：目的探討健脾養(yǎng)血祛風(fēng)方對脾虛型濕疹小鼠模型皮損處細胞間黏附分子(ICAM-1及VCAM-1)的影響及可能的作用機制。方法取健康雄性小鼠50只,隨機分為5組,即空白對照組、脾虛慢性濕疹模型組、脾虛慢性濕疹西藥治療組、脾虛慢性濕疹中藥治療高劑量組及低劑量組。中藥組以健脾養(yǎng)血祛風(fēng)方進行治療,西藥組以鹽酸左西替利嗪治療;采用免疫組化法檢測小鼠耳部病變組織ICAM-1及VCAM-1的水平。結(jié)果模型組小鼠耳部病變組織ICAM-1及VCAM-1均有不同程度的陽性表達;給藥后,脾虛慢性濕疹中藥治療高劑量組及西藥組小鼠耳部病變組織ICAM-1和VCAM-1的表達均較模型組顯著降低(P0.01)。中藥高劑量組與西藥組ICAM-1及VCAM-1的水平差異無統(tǒng)計學(xué)意義(P0.05)。結(jié)論健脾養(yǎng)血祛風(fēng)方能夠抑制脾虛型濕疹小鼠炎性細胞浸潤,減輕炎癥反應(yīng),其作用機制可能與抑制皮損處ICAM-1及VCAM-1的表達有關(guān)。
[Abstract]:Objective to investigate the effect and possible mechanism of invigorating spleen and nourishing blood and expelling wind on intercellular adhesion molecules (ICAM-1 and VCAM-1) in skin lesions of mice with spleen deficiency. Methods Fifty healthy male mice were randomly divided into 5 groups: blank control group, chronic eczema model group, chronic eczema treatment group, high dose group and low dose group. The traditional Chinese medicine group was treated with Jianpi Yangxue qufeng recipe, the western medicine group was treated with levcetirizine hydrochloride, and the levels of ICAM-1 and VCAM-1 in the ear lesions of mice were detected by immunohistochemical method. Results the expression of ICAM-1 and VCAM-1 in the ear lesions of the model group was significantly lower than that in the model group, and the expression of ICAM-1 and VCAM-1 in the high dose group and the western medicine group were significantly lower than that in the model group. There was no significant difference in ICAM-1 and VCAM-1 levels between high dose group and western medicine group (P 0.05). Conclusion Jianpi Yangxue qufeng recipe can inhibit the infiltration of inflammatory cells and alleviate inflammatory reaction in mice with spleen deficiency, and its mechanism may be related to the inhibition of the expression of ICAM-1 and VCAM-1 in skin lesions.
