濕疹納米乳膏急性毒性體外替代試驗(yàn)

【摘要】:目的:利用人KC系HaCaT細(xì)胞的體外細(xì)胞培養(yǎng),通過(guò)四甲基偶氮唑鹽比色法(MTT)法測(cè)定細(xì)胞的增殖活性,得到一組吸光度值(A值),根據(jù)吸光度值計(jì)算出細(xì)胞抑制率IC5。,來(lái)檢測(cè)濕疹納米乳膏的急性毒性。 方法:采用購(gòu)自湘雅中心實(shí)驗(yàn)室的人KC系HaCaT細(xì)胞進(jìn)行體外細(xì)胞傳代培養(yǎng)。濕疹納米乳膏稀釋成6個(gè)濃度組、及十二烷基硫酸鈉的陽(yáng)性對(duì)照組、陰性對(duì)照組、空白對(duì)照組共9組,分別作用于各組細(xì)胞一定時(shí)間后用MTT法測(cè)定細(xì)胞的增殖活性,得到一組吸光度值(OD值),重復(fù)實(shí)驗(yàn)3次,根據(jù)吸光度值計(jì)算出細(xì)胞抑制率,將細(xì)胞抑制率對(duì)濃度對(duì)數(shù)作圖,按作圖法用SPSS16.0程序計(jì)算出IC50。 結(jié)果:MTT實(shí)驗(yàn)表明:根據(jù)吸光度值得出濕疹納米乳膏6個(gè)濃度組的從低到高的細(xì)胞抑制率分別為86.017%、85.317%、83.94%、84.14%、87.584%、90.767%,將細(xì)胞抑制率對(duì)濃度對(duì)數(shù)作圖,按作圖法用SPSS16.0計(jì)算出:IC50為6.459mg/ml, IC50≥1.5mg/ml,說(shuō)明濕疹納米乳膏細(xì)胞毒性極小或無(wú)毒性。 結(jié)論:MTT法檢測(cè)濕疹納米乳膏對(duì)人KC系HaCaT細(xì)胞的細(xì)胞增殖的影響,最終實(shí)驗(yàn)結(jié)果為:濕疹納米乳膏的IC50值為6.459mg/ml, IC50≥1.5mg/ml,說(shuō)明濕疹納米乳膏細(xì)胞毒性極小或無(wú)毒性,濕疹納米乳膏經(jīng)皮吸收和短期應(yīng)用所產(chǎn)生的毒性反應(yīng)無(wú)或很小。濕疹納米乳膏應(yīng)用于臨床的急性毒性無(wú)或比較小,相對(duì)安全,可以用于臨床。
[Abstract]:Objective: to determine the proliferation activity of human KC HaCaT cells by MTT assay. A group of absorbance values were obtained and the cell inhibition rate (IC5. 5) was calculated according to the absorbance values to detect the acute toxicity of eczema nanometer cream. Methods: human KC HaCaT cells from Xiangya Central Laboratory were cultured in vitro. Eczema nanometer cream was diluted into 6 concentration groups and 9 groups of positive control group, negative control group and blank control group. After treated with sodium dodecyl sulfate for a certain time, the proliferative activity of the cells was measured by MTT method. A group of absorbance values and OD values were obtained and repeated experiments for 3 times. According to the absorbance values, the cell inhibition rate was calculated. The logarithm of cell inhibition rate against concentration was plotted, and the IC50 was calculated by SPSS16.0 program according to the drawing method. Results: according to the absorbency of eczema nanometer cream, the cell inhibition rates from low to high were 86.01717, 85.31717 and 83.94, respectively.
